非洲猪瘟病毒间接ELISA抗体检测方法的建立 被引量:19

Development of an ELISA Based on Synthetic Peptides for Detection of Antibodies Against the African Swine Fever Virus

摘要 非洲猪瘟(African swine fever,ASF)是一种急性、热性、高度接触性动物传染病,其病原为非洲猪瘟病毒(African swine fever virus,ASFV),临床上以高热、网状内皮系统出血和高死亡率为特征,易感猪群病死率高达100%。为了建立一种基于合成肽技术的非洲猪瘟病毒(African swine fever virus,ASFV)抗体血清学检测方法。本研究根据ASFV p30、p54和p72三种重要抗原设计3条合成肽,以此为包被抗原建立了ASFV间接ELISA抗体检测方法,并验证其敏感性、特异性、稳定性以及与进口试剂盒的符合率。结果显示,建立的间接ELISA方法的敏感性为91.4%,特异性为98.1%,批内和批间变异系数分别为3.7%~10.1%和4.7%~14.9%。与进口试剂盒比较,符合率为92.9%。结果表明该方法检测结果准确性高、且稳定性好,可检测ASFV特异性抗体。 African swine fever(ASF)is an acute,hot and highly contagious animal infectious disease caused by African swine fever virus(ASFV).It is clinically characterized by high fever,reticuloendothelial system hemorrhage and high mortality.The mortality rate of susceptible pigs is up to 100%.We wished to establish a serological method based on synthetic-peptide technology for detection of antibodies against the African swine fever virus(ASFV).Three synthetic peptides were designed based on three important antigens of the ASFV(p30,p54 and p72)and were used as coating antigens to detect antibodies against the ASFV.An ELISA system was optimized and the sensitivity,specificity and repeatability of this method were tested.The sensitivity and specificity was 91.4%and 98.1%,respectively.Variations in intra-and inter-assay repeatability was 3.7%–10.1%and 4.7%–14.9%,respectively.Comparing this method with that using a USA-imported kit showed a coincidence rate of 92.9%.Results showed that our method was accurate and stable,and could detect specific antibodies against the ASFV.
