摘要
目的探讨柴胡皂苷D对肝癌大鼠肿瘤生长及免疫功能的影响。方法按照随机数字表法将大鼠分为5组:正常组、模型组、对照组和低、高2个剂量实验组,各18只。大鼠给予二乙基亚硝胺溶液灌胃建立原发性肝癌模型。造模同时,低、高2个剂量实验组给予0.2,0.4 mg·mL^-1柴胡皂苷D溶液2 mL,qd,腹腔注射;对照组给予0.2 mg·mL^-1苦参碱溶液2 mL,qd,腹腔注射,连续给药16周。计算大鼠抑瘤率,用全自动生化分析仪测定血清谷丙转氨酶(GPT)水平,用流式细胞术测定外周血CD4+/CD8+。结果模型组、对照组和低、高2个剂量实验组的抑瘤率分别为0,(61.57±7.56)%,(28.49±3.12)%和(61.79±7.25)%;对照组和低、高2个剂量实验组与模型组相比,差异均有统计学意义(均P<0.05)。正常组、模型组、对照组和低、高2个剂量实验组的血清GPT水平分别为(60.74±6.86),(285.46±36.54),(164.63±20.45),(207.84±25.43)和(163.22±20.07)U·L^-1;这5组的CD4+/CD8+分别为2.44±0.33,0.94±0.12,1.72±0.21,1.29±0.16和1.75±0.24;模型组与正常组相比,差异均有统计学意义(均P<0.05);对照组和低、高2个剂量实验组与模型组相比,差异均有统计学意义(均P<0.05)。高剂量实验组和对照组效果类似。结论柴胡皂苷D能够剂量依赖性地抑制肝癌大鼠肿瘤生长,改善大鼠肝功能,调节免疫功能,提高抗肿瘤能力。
Objective To analyze the effect of Saikosaponin-D on tumor growth and immune function in liver cancer rats.Methods Male SD rats were selected,and randomly divided into five groups according to random number table:normal group,model group,control group,and experimental-L,-H groups,each with 18 rats.The primary liver cancer model was established by gavage of diethylnitrosamine in each group.At the same time of modeling experimental-L,-H groups were given 0.2,0.4 mg·mL^-1 Saikosaponin-D solution 2 mL,qd,intraperitoneal injection;control group was given 0.2 mg·mL^-1 matrine solution 2 mL,qd,intraperitoneal injection,for 16 weeks.The tumor inhibitory rate was calculated.Serum glutamic-pyruvic transaminase(GPT)level was measured by automatic biochemical analyzer.The ratios of CD4+/CD8+in peripheral blood was measured by flow cytometry.Results The tumor inhibitory rates in model group,control group and experimental-L,-H groups were respectively 0,(61.57±7.56)%,(28.49±3.12)%,(61.79±7.25)%;comparison between experimental-L,experimental-H group,control group and model group,the difference was significant(P<0.05).Serum GPT levels in normal group,model group,control group,and experimental-L,-H groups were respectively(60.74±6.86),(285.46±36.54),(164.63±20.45),(207.84±25.43),(163.22±20.07),U·L^-1;the ratio of CD4+/CD8+in the five groups were respectively 2.44±0.33,0.94±0.12,1.72±0.21,1.29±0.16,1.75±0.24;comparison between model group and normal group,the differences of the factors were significant(all P<0.05);comparison between control group,experimental-L,experimental-H group and model group,the differences of the factors were significant(all P<0.05).The high dose experimental group was similar to the control group on effect.Conclusion Saikosaponin-D can inhibit tumor growth in primary liver cancer rats in a dose-dependent manner,improve the liver function of rats,regulate immune function,and improve the anti-tumor ability.
