摘要
实验采用阳离子交换层析技术分离纯化植物乳杆菌素M1-UVs301。结果表明,植物乳杆菌素M1-UVs301分离纯化后在250~320 mL活性突出,精度纯化后在11~13 mL具有抑菌活性。纯化后植物乳杆菌素M1-UVs301比活力显著增加至26 321 AU·mg-1,蛋白凝胶上条带单一明显,在3.4 kDa左右抑菌活性显著。氨基酸及蛋白结构分析结果表明,该植物乳杆菌素M1-UVs301为一种疏水肽,二级结构中β-折叠含量较高,为其后续结构和特性的进一步分析提供基础。
In this study,plantaricin M1-UVs301 was separated and purified by cation exchange chromatography.The results showed that the activity of plantaricin was prominent at 250~320 mL after separation and purification,and the activity of plantaricin was significant at 11~13 mL after precision purification.After purification,the specific activity of plantaricin was significantly increased to 26321 AU·mg^-1,the single band on the protein gel was obvious,the antibacterial activity was significant at about 3.4 kDa.The analysis results of amino acid and protein structure showed that plantaricin M1-UVs301 is a hydrophobic peptide,the content ofβ-sheet is higher in the secondary structure,it provides the basis for the further analysis of its structure and characteristics.
