摘要
目的观察黄芪水溶性黄酮(AF)对S180荷瘤小鼠血清中VEGF及VEGFR-2水平的影响,从而探讨黄芪黄酮的抗肿瘤作用及其机制,为黄芪黄酮的进一步开发与应用提供实验依据。方法将研究对象随机分为正常对照组、模型对照组、环磷酰胺治疗组、黄芪黄酮实验组以及联合用药组。采用酶联免疫吸附分析法(ELISA)检测各组S180荷瘤小鼠血清中血管内皮生长因子(VEGF)及其受体2(VEGFR-2)的水平,检测结果用方差分析处理,并进行对比分析。结果模型对照组的S180荷瘤小鼠血清中VEGF和VEGFR-2水平升高;环磷酰胺治疗组的S180荷瘤小鼠血清中VEGF和VEGFR-2水平降低;黄芪黄酮实验组的S180荷瘤小鼠血清中VEGF和VEGFR-2水平也有所降低;联合用药组的S180荷瘤小鼠血清中VEGF和VEGFR-2水平明显降低。结论黄芪黄酮能够降低荷瘤小鼠体内VEGF和VEGFR-2水平,从而抑制肿瘤生长与转移。黄芪黄酮与环磷酰胺联合用药使荷瘤小鼠体内VEGF和VEGFR-2水平下降更加明显。
Objective To obsrve the effect of Astragalus Flavoneon serum VEGF and VEGFR-2 levels of S180 tumor-bearing mice,so as to provide experimental basis for the further development and application of Astragalus Flavone to explore the anti-tumor effect and mechanism of astragalus flavone.Methods The research subjects were randomly divided into normal control group,model control group,cyclophosphamide treatment group,Astragalus Flavone experimental group and combination therapy group.Serum levels of vascular endothelial growth factor(VEGF)and its receptor 2(VEGFR-2)in each group of S180 tumor-bearing mice were detected by enzyme-linked immunosorption assay(ELISA).Tested results of variance analysis with treatment,compared and analyzed.Results Serum levels of VEGF and VEGFR-2 were increased in S180 tumor-bearing mice in the model control group.Serum levels of VEGF and VEGFR-2 were decreased in S180 tumor-bearing mice treated with cyclophosphamide.Serum levels of VEGF and VEGFR-2 were also decreased in S180 tumor-bearing mice treated with Astragalus Flavone.Serum levels of VEGF and VEGFR-2 were significantly reduced in the combined group of S180 tumor bearing mice.Conclusion Astragalus Flavones can reduce VEGF and VEGFR-2 levels in tumor-bearing mice,thereby inhibiting tumor growth and metastasis.The combination of Astragalus Flavone and cyclophosphamide significantly reduced the levels of VEGF and VEGFR-2 in tumor-bearing mice.
