摘要
目的通过观察救顽汤对慢性湿疹大鼠模型血清中γ-干扰素(IFN-γ)、白介素-4(IL-4)与皮质酮(CORT)含量变化的影响,初步探讨救顽汤治疗慢性湿疹的作用机制。方法随机将60只雄性SD大鼠分为6组,每组10只,分别为空白组、模型组、阳性组、救顽汤低剂量组、中剂量组、高剂量组。除空白组外,各组大鼠运用2,4-二硝基氯苯(DNCB)复制慢性湿疹动物模型。模型建立后连续给药10 d,末次给药12 h后取样,比较各组间皮损程度以及血清中IFN-γ、IL-4与CORT含量变化。结果救顽汤高、中剂量能有效减轻慢性湿疹模型造成的皮损与炎症程度;能有效升高慢性湿疹模型降低的血清中IFN-γ、CORT的含量,降低慢性湿疹模型升高的血清中IL-4含量(P<0.01或P<0.05),低剂量组无差异(P>0.05)。结论救顽汤能有效调节慢性湿疹模型大鼠血清中异常表达的IFN-γ与IL-4水平,说明其作用机制与调节Th1/Th2平衡有关;救顽汤能有效升高慢性湿疹模型大鼠血清中的CORT含量,其作用机制还与调节糖皮质激素表达相关。
Objective To observing the effects of Jiuwan decoction on the changes of IFN-γ,IL-4 and Corticoid in serum of rats with chronic eczema,the mechanism of Jiuwan decoction in treating chronic eczema was preliminarily discussed.Methods 60 SD rats were randomly divided into 6 groups including blank group,model group,positive group,low-dose,mid-dose and high-dose groups with 10 rats in each group.The test groups were used to replicate the animal model of chronic eczema using 2,4-dinitrochlorobenzene(DNCB)except for the blank group.These rats were doses for 10 days after successful model establishment.The degree of skin lesions,spleen index and serum IFN-γ,Corticoid and IL-4 levels were compared among these groups.Results High and medium doses of Jiuwan decoction can effectively reduce the skin lesions and inflammation caused by chronic eczema model.It can effectively increase the content of IFN-γand CORT in the serum of the chronic eczema model and reduce the content of IL-4 in the serum of the chronic eczema model(P<0.01 or P<0.05).There was no difference in the low-dose group(P>0.05).Conclusion Jiuwan decoction can effectively regulate the abnormal expression of IFN-γand IL-4 levels in serum of rats with chronic eczema,indicating that its mechanism of action is related to the regulation of Th1/Th2 balance.Jiuwan decoction can effectively increase the CORT content in serum of rats with chronic eczema,and its mechanism is related to the regulation of glucocorticoid expression.
