摘要
采用直接水浴加热双水相法提取巴山雀舌茶树老叶中的黄酮,对分相盐进行了筛选,研究了乙醇体积分数、分相盐的用量、提取时间、提取温度变化对黄酮得率的影响.在单因素实验结果的基础上,以L9(34)正交设计实验得到了黄酮的最优提取条件:乙醇体积分数为70%,硫酸铵用量为5g/30mL,提取温度为60℃,提取时间为70min;此条件下进行验证实验,黄酮得率均值为6.61%,相对标准偏差为0.63%;提取产物对DPPH·清除率与其加入量呈良好的正相关关系,用浓度为0.0002mol/L的DPPH·进行实验,最大清除率为71.18%.双水相体系提取茶树老叶中的黄酮操作简便,对茶叶生产中副产物的综合利用有积极意义.
Dual aqueous phase extraction of flavonoids in old Bashan finch tea leaves was studied.By water-bath heating,effects of phylogenetic salt species,ethanol volume fraction,phylogenetic salt dosage,extraction time,and extraction temperature upon yield of flavonoids,were explored.By L 9(34)orthogonal tests,the optimal extraction conditions were acquired,namely,ethanol volume fraction of 70%,ammonium sulfate dosage of 5 g/30mL,extraction temperature of 60℃,and extraction time of 70min,under which the yield of flavonoids could reach 6.61%.The clearing rate of extract for DPPH·had good positive correlation with the added amount of extract.The clearing ability of extract to DPPH·was better than that of rutin.
