烯丙孕素-β-环糊精和羟丙基-β-环糊精包合物制备工艺优选 被引量:5

Optimization of Preparation Technology of Inclusion Compound of Altrenogest-β-cyclodextrin and Its Derivatives

摘要 研究烯丙孕素-β-环糊精包合物以及烯丙孕素-羟丙基-β-环糊精包合物制备工艺。采用正交试验法优选β-环糊精以及羟丙基-β-环糊精对烯丙孕素的包合工艺。选用圆二色谱法对包合物进行表征,并考察包合物的稳定性。以包合物中收率和包合率为指标,筛选出最佳包合条件为烯丙孕素与β-环糊精投料摩尔比为1∶3,65℃加热搅拌3 h;烯丙孕素与羟丙基-β-环糊精投料摩尔比为1∶6,55℃加热搅拌4 h。经圆二色谱法鉴定,已经形成包合物,并且具有一定稳定性。试验得出的制备工艺稳定可行。 To optimize the preparation procedure of inclusion compound of altrenogest-β-cyclodextrin and altrenogest-hydroxypropyl-β-cyclodextrin,the factors affecting inclusion compound were studied with orthogonal design.Characterization of inclusion complexes by circular dichroism and the stability of the inclusion complex was investigated.According to the yield and inclusion rate of the inclusion compound,the best inclusion conditions were selected.The molar ratio of altrenogest toβ-cyclodextrin is 1:3,and heating and stirring at 65℃for 3 h;The molar ratio of altrenogest toβ-cyclodextrin is 1:6,and heating and stirring at 55℃for 4 h.It was identified by circular dichroism that a clathrate was formed and had a certain stability.The preparation process obtained by the test is stable and feasible.
