摘要
为了探究美洲大蠊提取物Ento-A对大鼠急性胃黏膜损伤的保护作用。使用康复新液、西咪替丁、美洲大蠊提取物Ento-A高、低剂量灌胃大鼠,每天1次,连续8 d;末次给药后1 h,灌胃80%乙醇与20 mg/mL水杨酸钠溶液的等体积混合液建立急性胃黏膜损伤模型,90 min后观察胃黏膜损伤情况。酶联免疫法检测血清中PGE 2、COX-2、CRP的水平及胃组织匀浆中TGF-β1的水平。结果显示,与模型对照组比较,康复新液、西咪替丁及美洲大蠊提取物Ento-A能显著降低急性胃炎大鼠血清中COX-2、PGE 2、CRP的表达(P<0.01),并能显著提升胃组织匀浆中TGF-β1的水平(P<0.01)。表明美洲大蠊提取物Ento-A可明显减轻大鼠急性胃炎的损伤程度,可能与降低血清PGE 2、COX-2、CRP的水平及升高胃组织匀浆中TGF-β1的水平有关。
To investigate the protective effect of Periplaneta americana L Extract Ento-A on acute gastric mucosal injury in rats.The rats were treated with Kang Fu-xin,cimetidine and Ento-A high and low dose intragastric perfusion once a day for 8 days.An hour after the last administration,the model of acute gastric mucosal injury model was established by gavage of an equal volume mixture of 80%ethanol and 20 mg/mL sodium salicylate solution,and the gastric mucosal injury was observed after 90 min.The levels of PGE 2,COX-2 and CRP in serum and TGF-β1 in gastric homogenate were detected by enzyme linked immunosorbent assay(ELISA).The results showed that the compared with the model control group,Kang Fu-xin,cimetidine and extract of Periplaneta americana L Ento-A could significantly reduce the expression of COX-2,PGE 2 and CRP in serum of rats with acute gastritis(P<0.01),and significantly increased the level of TGF-β1 in gastric tissue homogenate(P<0.01).Indicated that Ento-A can significantly reduce the injury degree of acute gastritis in rats,which may be related to the decrease of serum PGE 2,COX-2 and CRP level and the increase of TGF-β1 level in gastric tissue homogenate.
