摘要
目的长链非编码RNAs(LncRNA)是通过基因组转录的方式辨认RNA物种。但其功能仍然未知,本研究旨在探究LncRNA调控心脏成纤维细胞活化增殖和纤维化的作用机制。方法在LncRNA数据库中鉴定出小鼠心脏高表达的LncRNA 1700020I14Rik。采用实时定量PCR(qRT-PCR)技术,检测小鼠心脏发育过程中LncRNA的表达。在横断主动脉条带介导的小鼠心肌肥大中检测其表达,然后在分离的心肌成纤维细胞中过量表达LncRNA以确定其对心肌成纤维细胞增殖的影响。结果在哺乳动物中LncRNA的功能是保守的。其在小鼠心脏中选择性表达,并在成年期高度表达,这一功能在肥厚型心脏的成纤维细胞中受到抑制。在心脏成纤维细胞中,LncRNA抑制心肌成纤维细胞增殖和胞外基质蛋白的积累。结论LncRNA 1700020I14Rik是一种心肌富集型LncRNA,在应激性心肌纤维化中表达下调功能。LncRNA 1700020I14Rik有抑制心肌成纤维细胞增殖和胶原合成及抑制心肌纤维化的作用。
Objective Long non-coding RNAs(LncRNAs)are newly identified RNA species by genome-wide transcriptome sequencing.Although numerous LncRNAs have been found,the functions remain largely unknown.The purpose of this study is to explore its functions.Methods A murine cardiac highly expressed LncRNA 1700020I14Rik was identified in LncRNA database.The expression of the LncRNA during mouse heart development process was detected by using real-time quantitative PCR.Its expression was also examined in a transverse aortic banding-mediated myocardial hypertrophy in mouse.Then,the LncRNA was overexpressed in isolated cardiac fibroblasts to determine its effects on cardiac fibroblast proliferation.Results The LncRNA is conserved among mammals.In mouse,it is selectively expressed in hearts and highly expressed in adult stages,which is inhibited in cardiac fibroblasts in hypertrophic hearts.In cardiac fibroblasts,it inhibits cardiac fibroblasts proliferation and extracelluar matrix protein accumulations.Conclusion LncRNA 1700020I14Rik is a cardiac-enriched LncRNA,which is down-regulated in stressed hearts associated with fibrosis.LncRNA 1700020I14Rik inhibits cardiac fibroblast proliferation and collagen synthesis,indicating its functions in repressing cardiac fibrosis.
