摘要
目的采用高效液相色谱(HPLC)法测定广西省内12种不同产地和9种炮制品的三叉苦茎中吴茱萸春碱成分的含量,为评价广西三叉苦茎的质量优劣。方法采用Anilent ZORBAX SB-C18(4.6 mm×250 mm,5-Micron)色谱柱;流动相:以乙腈为B,纯水为D,进行梯度洗脱;柱温:35℃;流速:1 mL/min;进样量:5μL;检测波长:250 nm。并分别对不同产地和不同炮制品三叉苦茎中的吴茱萸春碱成分的含量进行了分析。结果建立了三叉苦茎中吴茱萸春碱成分的含量测定方法,该成分的质量浓度和峰面积均呈良好的线性关系;方法的精密度、稳定性和重复性均RSD<2.36%;平均加样回收率为105.09%;RSD值为5.12%。不同炮制品吴茱萸春碱含量:蜜炙>炒黄>烘干>酒炙>晒干>盐炙>炒焦>醋炙>阴干;不同产地吴茱萸春碱含量:S9>S10>S7>S2>S12>S3>S8>S6>S5>S4>S1>S11。结论建立了三叉苦茎中吴茱萸春碱成分的定量分析方法,为三叉苦茎的质量控制提供参考依据。
Objective In order to evaluate the quality of trigeminal bitter stem in Melicope pteleifolia in Guangxi,the contents of evodiamine in 12 different producing areas and 9 processed products were determined by high performance liquid chromatography(HPLC).Methods An Anilent ZORBAX SB-C18(4.6 mm×250 mm,5-Micron)column was used.Mobile phase consisted of acetonitrile(B)-water(D).The flow rate was set at 1 mL/min and the column temperature was set at 35℃.Injection volume was 5μL and detection wavelength was 250 nm.The contents of evolitrine in different producing areas and processed products were analyzed.Results A method for the determination of evolitrine in the Melicope pteleifolia was established.The quality concentration and peak area of the component showed a good linear relationship.The RSD of the precision,stability and repeatability of the method was less than 2.36%.The average recovery rate was 105.09%,and the RSD value was 5.12%.The content of evolitrine in different processed products was as follows:honey roasted>stir fried yellow>dried>wine roasted>sun dried>salt roasted>stir fried>vinegar roasted>dried in the shade.The content of Evolitrine in different producing areas was as follows:S9>S10>S7>S2>S12>S3>S8>S6>S5>S4>S1>S11.Conclusion The quantitative analysis method of evolitrine in Melicope pteleifolia was established,which could provide reference for the quality control of Melicope pteleifolia.
