高海拔地区不同年龄组慢性牙周患者群TNF-α-308和IL-Ⅰ-899基因型研究

TNF-α-308 and IL-Ⅰ-899 Genotype Study in Different Age Groups of Chronic Periodontal Disease at High Altitude Area

摘要 目的:探讨肿瘤坏死因子(TNF-α)-308位点基因多态性和白细胞介素Ⅰ(IL-Ⅰ)-899位点基因多态性与青海高海拔地区不同年龄组人群慢性牙周炎易感性的基因层面相关性研究。方法:选取慢性牙周炎患者128例,按年龄分为(19~34)岁、(35~44)岁、(45~54)岁、(55~65)岁4个组。采集患者颊黏膜拭子,采用聚合酶链反应限制性片段长度多态性(PCR-RFLP)基因分析法,测定TNF-α-308位点基因型和IL-Ⅰ-899位点基因型等位基因分布。结果:TNF-α-308位点3种基因型和G、C等位基因频率在4个年龄组间分布差异有统计学意义(P<0.05)。等位基因C的表达明显高于G的表达,等位基因型CC纯合子(45~54)岁组明显高于(35~44)岁组,差异有统计学意义(P<0.05)。IL-Ⅰ-899位点Ⅰ/Ⅱ多态性分布、IL-Ⅰ-899位点3种基因型和Ⅰ、Ⅱ等位基因频率在4个年龄组间分布行χ^2检验,差异无统计学意义(P>0.05)。4个年龄组等位基因频率Ⅰ的表达明显高于Ⅱ的表达,均以纯合子Ⅰ/Ⅰ基因型分布为主,高于Ⅰ/Ⅱ杂合子。结论:高海拔地区慢性牙周病患者中TNF-α-308位点基因型变化与年龄有相关性,IL-Ⅰ-899位点基因变化与年龄组无相关性。 Objective:This study is to investigate the correlation between tumor necrosis factor(Tumor Necrosis Factor-α,TNF-α)-308 locus gene polymorphism and InterleukinⅠ(InterleukinⅠ,IL-Ⅰ)-899 locus gene polymorphism and chronic periodontitis susceptibility in different age groups at high altitude area.Methods:128 cases of patients with chronic periodontitis grouping by age were divided into four groups of 19-34,35-44,45-54,55-65 years old.The mucous swabs of the buccal mucosa from the patients were collected.Polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP)gene analysis was used to detect allele distribution in TNF-α-308 and IL-Ⅰ-899 loci genotype.Results:Chi square test was used respectively,to test the distribution of 3 genotype of TNF-α-308 locus and G、C allele frequencies in 4 age groups of chronic periodontitis patients in Qinghai area.Chi square test showed significant difference(P<0.05).The expression of allele C was significantly higher than that of G,and the allelic CC homozygote in 45-54 year old group was significantly higher than that in the 35-44 year old group(P<0.05).Chi square test was used respectively,to detect the distribution ofⅠ/Ⅱpolymorphism in IL-Ⅰ-899 locus,IL-Ⅰ-899 loci 3 genotype and allele frequencies ofⅠ/Ⅱat 4 age groups of chronic periodontitis patients in Qinghai area.There was no significant difference in the Chi square test(P>0.05).The expression of alleleⅠwas significantly higher than that ofⅡ,which was mainly based on the homozygousⅠ/Ⅰgenotype.TheⅠ/Ⅱheterozygote in the four groups was the least.TheⅠ/Ⅰgenotype was significantly higher thanⅠ/Ⅱ,which was tested by Chi square in the same age group.The difference between each group was significant difference(P<0.01).Conclusions:The genotype of TNF-α-308 and IL-Ⅰ-899 in patients with chronic periodontal disease were correlated with ages.
