摘要
目的建立复方苦黄凝胶剂的高效液相色谱(HPLC)指纹图谱。方法采用Agilent SB-C18(4.6×150 mm,5μm)色谱柱,以乙腈-0.5%磷酸为流动相,流速1.0 mL/min,梯度洗脱,检测波长220 nm,柱温30℃,洗脱时间为90 min。指纹图谱相似度评价采用中药色谱指纹图谱相似度评价系统(2004A版)。结果建立了复方苦黄凝胶剂的HPLC指纹图谱,确立了22个共有峰,4个峰归属到各药味,其中峰1归属为苦参,峰15归属为黄柏,峰20归属为蛇床子,峰11归属为防风。10批样品指纹图谱的整体相似度均在0.9以上。结论所建立的复方苦黄凝胶剂指纹图谱能够为整体控制制剂质量提供参考。
Objective To establish the high performance liquid chromatograhy(HPLC)fingerprint of compound Kuhuang gel.Methods The column was Agilent SB-C18(4.6×150 mm,5μm)and the mobile phase was acetonitrile-0.5%phosphoric acid solution with gradient elution at a flow rate of 1.0 mL/min.The column temperature was 30℃and the detection wavelength was 220 nm.The elution time was 90 minutes.The fingerprint similarity evaluation was performed by the traditional Chinese medicine(TCM)fingerprint similarity evaluation system of 2004 edition.Results The HPLC fingerprint of the compound Kuhuang gel was established;22 common peaks were established;4 peaks were assigned to each medicinal flavor.Peak 1 was assigned to Sophora flavescens,peak 15 was assigned to Phellodendron chinense,and peak 20 was assigned to Cnidium,peak 11 attributed to wind protection.The overall similarity of the fingerprints of the 10 batches of samples was above 0.9.Conclusion The fingerprint of the compound Kuhuang gel could provide a reference for the overall quality control of the preparation.
