鸡源沙门氏菌(Salmonella)致病基因与致病性的相关性研究 被引量:9

Analysis on the Relationship between Virulence Genes and Pathogenicity of Chicken Salmonella Isolates

摘要 为分析沙门氏菌分离株致病基因与致病性之间的关系,采集经分离鉴定的55株鸡源沙门氏菌,应用PCR方法检测毒力基因,通过小鼠致病试验检测致病性。结果显示,所检测的质粒毒力基因spvR、spvA、spvB、spvC、spvD,检出率均在60%以上;所检测的毒力岛毒力基因invH、sipA、sipB、sopA、sopD、avrA、hilA、iacP、prgK、ssaB、ssaQ、sifA、sseL、ssrA、ttrB、mgtC、misL、rmbA、rhuM、sugR、orf319、siiD、siiE、spi4H、sopB和pipC中,ttrB检出率最高,为92.73%,sifA检出率最低,为5.45%。55株分离株对小鼠均具有致病性,致死率为20%~100%;随机测定其中10株的半数致死量(LD50),为4×107~3.18×108CFU/mL,毒力基因检出数量越多的分离株,其LD50值越小,毒力越强。本研究结果揭示,鸡源致病性沙门氏菌分离株普遍携带毒力基因,其毒力基因与致病性之间呈现正相关,为深入研究沙门氏菌的致病机理提供基础数据。 To explore the relationship of virulence genes and pathogenicity of Salmonella isolates, virulence genes were detected by PCR from 55 chicken Salmonella isolates, and their pathogenicity was determined by challenge experiment in mice. The results showed that the plasmid virulence genes which included spvR, spvA, spvB, spvC and spvD were detected in more than 60% isolates. Among the pathogenicity island virulence genes, which included invH, sipA, sipB, sopA, sopD, avrA, hilA, iacP, prgK, ssaB, ssaQ, sifA, sseL, ssrA, ttrB, mgtC, misL, rmbA,rhuM, sugR, orf319, siiD, siiE, spi4H, sopB and pipC, ttrB was the most common in 92.73% isolates, while sifA was the least with 5.45% positive rate. All the 55 Salmonella isolates were pathogenic to mice with 20%~100%mortality rate. The median lethal dose(LD50) of 10 randomly selected isolates was from 4×107 CFU/mL to 3.18×108 CFU/mL, showing that the more virulence genes harbored, the less LD50 value was determined. The results in this study indicated that the clinical Salmonella isolates commonly harbored virulence genes and there existed positive relationship between virulence genes and bacterial pathogenicity, which supported the basic data for further research on the pathogenic mechanism of Salmonella.
