摘要
为了寻找能抑制肿瘤细胞增殖的药物,本研究探讨了不同浓度白花蛇舌草(Hedyotis diffusa)对人胃癌MKN-45细胞的影响,使用不同浓度的白花蛇舌草处理人胃癌MKN-45细胞24 h、48 h、72 h,共聚焦显微镜下观察细胞形态,流式细胞术检测细胞周期,qRT-PCR和Western blotting检测CCNB1和CCND1的表达水平。结果表明,在一定浓度范围内,白花蛇舌草可抑制人胃癌MKN-45细胞增殖且呈剂量时间依赖性;白花蛇舌草处理人胃癌MKN-45细胞24 h、48 h、72 h后的IC50值分别为111.759μg/mL、26.878μg/mL、16.179μg/mL;随着白花蛇舌草的浓度增加,显微镜视野下细胞数量减少;G1期细胞减少,S期细胞增多;CCNB1和CCND1基因表达水平显著降低。白花蛇舌草能能抑制人胃癌MKN-45细胞增殖,阻滞细胞于S期,降低CCNB1和CCND1基因的表达。
In order to find a drug that can inhibit the proliferation of human gastric cancer cells,the effects of different concentrations of Hedyotis diffusa on human gastric cancer MKN-45 cells were studied in this paper.Human gastric cancer MKN-45 cells were treated with different concentrations of Hedyotis diffusa for 24 h,48 h and 72 h.Cell morphology was observed under confocal microscope,flow cytometry was used to detect cell cycle,q RT-PCR and Western blotting was used to detect the expression of CCNB1 and CCND1 genes.The results showed that the proliferation of human gastric cancer MKN-45 cells could be inhibited in a dose-dependent manner in a certain concentration range.The IC50 values of gastric cancer MKN-45 cells were 111.759μg/mL,26.878μg/mL and 16.179μg/mL for 24 h,48 h,72 h that treated with Hedyotis diffusa,the number of cells decreased under microscope;G1 phase cell reduced,S phase cells increased;The expression level of CCNB1 and CCND1 genes decreased significantly.Conclusion:Hedyotis diffusa can inhibit the proliferation of human gastric cancer MKN-45 cells,block the cells in S phase,and decrease the expression of CCNB1 and CCND1 genes.
