眼子菜茎的组织培养及快速繁殖体系建立 被引量:1

摘要 为眼子菜的药用开发提供种质资源和技术支撑,以眼子菜茎为试验材料,研究不同浓度6-糠基氨基嘌呤(KT)、萘乙酸(NAA)、6-苄基氨基嘌呤(6-BA)、吲哚丁酸(IBA)、无机盐和糖对眼子菜茎段出芽数、叶片数、根状茎节数及生根数的影响。结果表明:眼子菜芽的最佳诱导培养基为MS+KT 0.05 mg/L+NAA 0.1 mg/L,该条件下的出芽数为10个,大部分形成不定芽,诱导能力很好;最佳增殖培养基为MS+6-BA 1 mg/L+NAA 0.05 mg/L,其出芽数、叶片数和根状茎数分别为5.3个、9张和8.3节;最佳生根培养基为1/2MS+IBA 0.5 mg/L+无菌水,其出芽数、叶片数、根状茎节数和生根数分别为5.7个、15.0张、8.7节和13.7根;组培苗在模拟水团环境中的移栽存活率为98%。 The effects of 6-furfuryl aminopurine(KT),naphthylacetic acid(NAA),6-benzylaminopurine(6-BA),indolebutyric acid(IBA),inorganic salt and sugar with different concentrations on budding number,leaf number,rhizome node number and rooting number of Potamogeton distinctus stems are studied to provide germplasm resource and technological support for medicinal development of P.distinctus.Result:The optimal inducing medium for P.distinctus bud is MS+0.05 mg/L KT+0.1 mg/L NAA,the budding number is 10,and most of them forms adventitious bud,and the inducibility is very good under the optimal inducing medium.The optimal proliferation medium for P.distinctus bud is MS+1 mg/L 6-BA+0.05 mg/L NAA,the budding number,leaf number and rhizome node number is 5.3,9 and 8.3 respectively.The optimal rooting medium for P.distinctus bud is 1/2MS+0.5 mg/L IBA+sterile water,and the budding number,leaf number,rhizome node number and rooting number is 5.7,15.0,8.7 and 13.7 respectively.The survival rate of tissue culture seedling is 98%under the environment of simulated paddy field.
