摘要
[目的]樱花根癌病近年对樱花产业的威胁日益严重,生产上亟需有效的防治措施。本研究旨在探讨瓦雷兹芽孢杆菌JK-XZ8的抑菌物质特性,同时测定其对樱花根癌病的田间防治效果。[方法]在平板拮抗测定菌株拮抗能力,通过酸沉淀提取该菌株的抑菌粗提物;利用PCR技术检测该菌是否含有推测的抑菌物质合成相关基因,测定该抑菌粗提物在不同条件下的稳定性;采用根部蘸浆法测定瓦雷兹芽孢杆菌(Bacillus velezensis)JK-XZ8对田间樱花苗木的生防效果。[结果]瓦雷兹芽孢杆菌JK-XZ8对根癌土壤杆菌存在拮抗能力且可产生抑菌物质;经PCR检测该菌株含有脂肽类基因bmyB,ituD,fenD和聚酮类化合物基因PK1、PK2、PK3等抑菌物质合成相关基因。此外粗提得到的抑菌物质,在高温、酸、碱、蛋白酶K、3 h内的紫外线的处理条件下,仍保持较高活性。田间试验表明菌株JK-XZ8对樱花根癌病的防治效果可达50%,优于市场上使用的K84生物菌肥(24%)。[结论]瓦雷兹芽孢杆菌可以产生稳定的抑菌物质拮抗病原菌,是一株可用于樱花根癌病防治的有效生防菌株。
[objective]Cherry blossom crown gall has become a serious threat to the cherry blossom industry in recent years,and effective control measures are urgently needed in production.Present study aimed to explore the antibacterial properties of Bacillus velezensis JK-XZ8,and determine its field control effect on cherry blossom crown gall.[Methods]Plate antagonism method was employed to determine the antagonistic ability of the strain.The crude bacteriostatic extract of the strain was extracted by acid precipitation,and the bio-control effects of B.velezensis JK-XZ8 on wild cherry tree seedlings was determined by root dipping method.The stability of the crude extract was determined under different conditions.PCR technology was used to detect the potential genes in the bacteria that relate to antibacterial substance synthesis.[Results]B.velezensis JK-XZ8 had antagonistic ability to against pathogenic bacteria with produced bacteriostatic substances.PCR testing revealed that this strain contained lipopeptide genes bmyB,ituD,fenD and polyketide genes PK1,PK2,PK3,the antibacterial substance synthesis related genes.In addition,the extracted antibacterial substances maintained high activity and stability under the treatment conditions of high temperature,acid and alkali,and were not degraded by proteinase K and ultraviolet light within three hours.Field trials have shown that the effects of strain JK-XZ8 on cherry root cancer reached up to 50%,which was better than the K84 biological bacterial fertilizer(24%)used in the market.[Conclusion]B.velezensis JK-XZ8 produced stable bacteriostatic substances to antagonize pathogenic bacteria,and it is an effective bio-control strain that can be used to control the flower cherry trees crown gall.
