基于NIR技术建立同时测定白芍饮片5种成分的定量分析模型 被引量:6

Establishing Quantitative Analysis Models for Simultaneous Determination of Five Components in Baishao(Paeoniae Radix)Based on NIR Technology

摘要 目的采用近红外光谱技术建立同时测定白芍饮片中芍药苷、芍药内酯苷、苯甲酰芍药苷、没食子酸和β-PGG 5种化学成分的定量分析模型。方法以109份白芍饮片作为研究对象,采用近红外漫反射光谱技术采集样本光谱信息,以HPLC测定的含量作为参考值,运用偏最小二乘法,结合SNV/MSV+导数+N-D平滑等预处理方法,分别建立白芍总苷成分(芍药苷、芍药内酯苷、苯甲酰芍药苷)、没食子酸和β-PGG 3个定量分析模型,并对模型进行验证。结果配对t检验及含量偏差结果表明5种化学成分的NIR预测值与参考值无显著差异,预测结果良好;其定量模型校正集相关系数R分别为0.91929、0.94610、0.94577、0.99125和0.93529,校正均方差分别为0.179、0.230、0.304、0.473和0.0367,预测均方差分别为0.148、0.243、0.160、0.472和0.0283。结论白芍饮片多指标定量分析模型能够快速的检测出上述5种化学成分的含量,结果准确可靠;近红外技术可以用于白芍饮片多成分定量分析模型的建立,为白芍饮片的质量控制提供新的分析方法。 Objective To establish the quantitative model for simultaneous determination of 5 chemical components of Baishao(Paeoniae Alba)pieces,including paeoniflorin,albiflorin,benzoyl paeoniflorin,gallic acid andβ-PGG by using near infrared spectroscopy.Method Taking 109 parts of Baishao(Paeoniae Alba)pieces as the research objects,the sample spectral information was collected by near-infrared diffuse reflectance spectroscopy technique to establish quantitative analysis model of total glycosides of radix paeoniae alba(paeoniflorin,albiflorin,benzoyl paeoniflorin),gallic acid andβ-PGG respectively by using partial least squares method combined with pretreatment methods including SNV/MSV,derivative and N-D smoothing,and validate the models.Result The results of paired t-test and content deviation showed that the NIR predictive value of 5 chemical constituents was not significantly different from the reference value and the results were satisfactory.The correlation coefficient(R)of the quantitative model calibration set was 0.91929,0.94610,0.94577,0.99125 and 0.93529.Root mean square error of calibration was 0.179,0.230,0.304,0.473 and 0.0367.Root mean square error of prediction was 0.148,0.243,0.160,0.472 and 0.0283.Conclusion The multi-index quantitative analysis model of Baishao(Paeoniae Alba)pieces can quickly detect the content of the 5 chemical constituents and the results were accurate and reliable.The near infrared technology can be used to establish a multi-component quantitative analysis model of Baishao(Paeoniae Alba)pieces and provide a new analytical method for quality control of Baishao(Paeoniae Alba)pieces.
