摘要
目的观察抗疏强骨合剂对去势骨质疏松模型大鼠骨组织中自噬相关蛋白LC3-Ⅱ、Beclin1、p62表达的影响,探讨抗疏强骨合剂治疗骨质疏松的可能作用机制。方法将120只雌性SD大鼠随机分为空白组、模型组、抗疏强骨合剂组、阿仑膦酸钠组,每组30只。除空白组外,其余各组大鼠采用双侧卵巢切除法建立骨质疏松模型,造模后抗疏强骨合剂组给予抗疏强骨合剂灌胃3个月,阿仑膦酸钠组给予阿仑膦酸钠水溶液灌胃3个月。观察各组大鼠的一般情况、骨组织病理学改变以及骨组织中自噬相关蛋白LC3-Ⅱ、Beclin1、p62表达情况。结果模型组大鼠精神状况较差,毛发颜色晦暗没有光泽,体质量下降显著,偶有毛发脱落;阿仑膦酸钠组、抗疏强骨合剂组大鼠精神状况尚可,毛发颜色较空白组稍差,体质量轻微下降,但抗疏强骨合剂组大鼠的一般情况更接近于空白组。HE染色示模型组大鼠骨髓腔内大量脂肪细胞增生,部分脂肪细胞核消失、碎裂,空骨陷窝数明显增多,骨小梁稀疏、结构紊乱;抗疏强骨合剂组大鼠骨髓腔脂肪细胞形态基本正常,偶有部分细胞增生,软骨下及骨髓腔骨小梁粗大、结构完整,空泡陷窝较少见,接近空白组;阿仑膦酸钠组大鼠的变化与抗疏强骨合剂组相似,但空泡陷窝较抗疏强骨合剂组多。与空白组比较,模型组大鼠骨组织中LC3-Ⅱ、Beclin1的相对灰度值升高(P均<0.05),p62相对灰度值降低(P<0.05);与模型组比较,阿仑膦酸钠组和抗疏强骨合剂组大鼠骨组织中LC3-Ⅱ、Beclin1的相对灰度值降低(P均<0.05),p62相对灰度值升高(P均<0.05);抗疏强骨合剂组、阿仑膦酸钠组、空白组各指标比较差异均无统计学意义(P均>0.05)。结论抗疏强骨合剂可能通过降低骨组织中自噬相关蛋白的含量,抑制自噬起到治疗骨质疏松症的作用。
Objective It is to observe the effect of Kangshu Qianggu mixture(KQM)on the expression of autophagy-related proteins LC3-Ⅱ,Beclin1 and p62 in the bone tissue of ovariectomized osteoporosis model rats,and explore the possible mechanism of this medicine in the treatment of osteoporosis.Methods 120 SD rats were randomly divided into blank group,model group,KQM group and alendronate group,with 30 rats in each group.Except for the blank group,the rats in the other groups were used to establish osteoporosis models by bilateral ovariectomy.After modeling,the KQM group was treated with Kangshu Qianggu mixture by gavage for 3 months,and the alendronate group was treated with alendronate solution by gavage for 3 months.The general condition,bone histopathological changes and the expression of autophagy-related proteins LC3-Ⅱ,Beclin1,p62 in the bone tissue of rats in each group were observed.Results The mental state of the rats in the model group was poor,their hair color was dull and lustreless,body weight decreased significantly,and the hair fell off occasionally;the rats in the alendronate group and KQM group were mentally healthy,and their hair color was slightly worse than that in the blank group,body weight decreased slightly,but the general situation of the rats in KQM group was closer to that of the blank group.HE staining showed that a large number of adipocytes proliferated in the bone marrow cavity of rats in the model group,some of the adipocyte nuclei disappeared and fragmented,the number of empty lacunae increased significantly,the bone trabecula was sparse,and the structure was disordered;the morphology of adipocytes in the bone marrow cavity of rats in the KQM group was basically normal,with some cell proliferation occasionally,the subchondral and bone marrow cavity trabeculae was thick,and the structure was complete with rare vacuolar lacunae,it was close to the blank group;the changes of rats in the alendronate group were similar to those in the KQM group,but its vacuoles lacunae was more than the KQM group.Compared with the blank group,the relative gray value of LC3-II and Beclin1 in the bone tissue of the model group was increased(P<0.05),and the relative gray value of p62 was decreased(P<0.05).Compared with the model group,the relative gray value of LC3-II and Beclin1 in the bone tissue of the alendronate group and KQM group was decreased(P<0.05),and the relative gray value of p62 was increased(P<0.05).There was no significant difference in the values among alendronate group and KQM group,blank group(P>0.05).Conclusion Kangshu Qiang bone mixture may reduce the content of autophagy-related proteins in bone tissue and inhibit autophagy to treat osteoporosis.
