老年心力衰竭合并肺部感染患者血清lncRNA NEAT1、miR-129表达及其临床意义 被引量:6

Expressions and Clinical Significance of Serum LncRNA NEAT1 and miR-129 in Elderly Patients with Heart Failure and Pulmonary Infection

摘要 目的探讨老年心力衰竭合并肺部感染患者血清长链非编码RNA(lncRNA)核富含丰富的转录本1(NEAT1)、微RNA-129(miR-129)表达水平变化及其临床意义。方法选取2014年9月至2019年4月保定市第一医院老年病科收治的260例老年心力衰竭患者为研究对象,根据是否发生肺部感染分为对照组(未发生肺部感染,n=136)和观察组(发生肺部感染,n=124)。采用实时荧光定量聚合酶链反应检测患者血清lncRNA NEAT1、miR-129表达水平,采用Pearson法分析血清lncRNA NEAT1、miR-129表达与血清脑钠肽(BNP)、降钙素原(PCT)水平的相关性。结果观察组血清BNP、PCT水平显著高于对照组(P<0.05);观察组血清lncRNA NEAT1表达水平明显高于对照组(3.96±1.01比1.09±0.32),观察组血清miR-129表达水平明显低于对照组(1.05±0.34比1.32±0.42)(P<0.05);其中心功能Ⅲ级、Ⅳ级组血清lncRNA NEAT1表达水平显著高于心功能Ⅱ级组(3.79±1.24、4.02±1.43比1.08±0.35)(P<0.05),心功能Ⅳ级组血清miR-129表达水平显著低于心功能Ⅱ级和Ⅲ级组(0.95±0.34比1.43±0.42、1.26±0.37)(P<0.05);Pearson法分析结果显示,观察组血清lncRNA NEAT1与BNP、PCT水平均呈正相关(r=0.472,P<0.001;r=0.464,P<0.001),血清miR-129与BNP、PCT水平均呈负相关(r=-0.441,P=0.001;r=-0.507,P<0.001)。结论老年心力衰竭合并肺部感染患者血清lncRNA NEAT1表达水平升高,血清miR-129表达水平降低,可能成为老年心力衰竭合并肺部感染的血清标志物。 Objective To explore the changes and significance of serum long non-coding RNA nuclear-enriched abundant transcript 1(lncRNA NEAT1)and microRNA-129(miR-129)expression levels in elderly patients with heart failure and pulmonary infection.Methods From Sep.2014 to Apr.2019,260 elderly patients with heart failure admitted to Department of Geriatrics of Baoding First Hospital were included in the study.Among them,124 patients with pulmonary infection were divided into an observation group and 136 patients without infection into a control group.The serum expression levels of lncRNA NEAT1 and miR-129 of the two groups were detected by real time quantitative polymerase chain reaction,and Pearson method was used to analyze the correlations between the expressions of serum lncRNA NEAT1,miR-129 and the levels of serum brain natriuretic peptide(BNP),procalcitonin(PCT).Results The levels of serum BNP and PCT in the observation group were significantly higher than those in the control group(P<0.05);the expression level of serum lncRNA NEAT1 in the observation group was significantly higher than that in the control group(3.96±1.01 vs 1.09±0.32),the expression level of serum miR-129 in the observation group was significantly lower than that in the control group(1.05±0.34 vs 1.32±0.42)(P<0.05);the level of serum lncRNA NEAT1 in thecardic functionⅢandⅣgroup was significantly higher than that of cardiac functionⅡgroup(3.79±1.24,4.02±1.43 vs 1.08±0.35)(P<0.05),and the expression level of serum miR-129 in cardiac functionⅣgroup was significantly lower than that in cardiac functionⅡandⅢgroup(0.95±0.34 vs 1.43±0.42,1.26±0.37)(P<0.05);the results of Pearson analysis showed that the serum lncRNA NEAT1 was positively correlated with BNP and PCT level(r=0.472,P<0.001;r=0.464,P<0.001),and the serum miR-129 was negatively correlated with BNP and PCT level(r=-0.441,P=0.001;r=-0.507,P<0.001).Conclusion The expression level of serum lncRNA NEAT1 in the elderly patients with heart failure and pulmonary infection is increased,and the expression level of serum miR-129 is decreased,which may be the serum markers of the elderly patients with heart failure and pulmonary infection.
