电针对慢性应激抑郁大鼠中缝核脑区P11-tPA-BDNF通路的影响 被引量:2

Effect of Electro-Acupuncture on P11-tPA-BDNF Pathway in the Raphe Nuclei of Rats with Chronic Stress Depression

摘要 目的:观察电针对慢性应激抑郁模型大鼠中缝核中P11、tPA及BDNF的影响,进一步研究电针治疗抑郁症可能的通路。方法:将24只健康雄性SD大鼠随机分为正常组、模型组、药物组和电针组,每组6只。除正常组外,其余各组均孤养,并接受慢性不可预知应激刺激造模。各组大鼠分别于实验前1 d和实验的第28天进行体质量检测和糖水偏好实验,观察大鼠的行为学变化。实验第28天断头取脑,采用免疫荧光染色技术检测中缝核中三者的表达情况;逆转录酶聚合酶链反应检测中缝核中P11、tPA、BDNF mRNA的含量。结果:模型组大鼠的行为学指标明显低于正常组(P<0.05),药物组和电针组大鼠行为学指标均明显高于模型组(P<0.05);模型组免疫荧光染色结果显示模型组三者在中缝核的表达明显低于正常组,药物组和电针组明显高于模型组;模型组中缝核P11、tPA、BDNF的mRNA含量低于正常组(P<0.01),药物组和电针组则显著高于模型组(P<0.01)。结论:电针有明显的抗抑郁效果,可以改善慢性应激抑郁大鼠的行为;电针可以增加慢性应激抑郁大鼠模型中缝核P11、tPA、BDNF mRNA的含量,提高其表达量,电针的这种抗抑郁作用可能是通过影响P11-tPA-BDNF通路发挥作用的。 Objective:To observe the effects of electro-acupuncture on P11,tPA and BDNF in the raphe nuclei of rats with chronic stress depression,and to further study the possible pathways of electro-acupuncture in treating depression.Methods:24 healthy male SD rats were randomly divided into the normal group,the model group,the medication group and the electro-acupuncture group,with 6 rats in each group.Except the normal group,the rats in the other groups were isolated and subjected to chronic unpredictable stress stimulation to establish the model.The body mass test and the syrup preference test were performed 1 day before the experiment and on the 28th day of the experiment,and the behavioral changes of the rats were observed.On the 28th day of the experiment,the brain sample was collected to detect the contents of P11,tPA and BDNF in the raphe nuclei by immunofluorescence staining;the mRNA contents of P11,tPA and BDNF in the raphe nuclei were detected by reverse transcriptase polymerase chain reaction.Results:The behavioral indexes of the model group were significantly lower than those of the normal group(P<0.05).The behavioral indexes in the medication group and the electro-acupuncture group were significantly higher than those in the model group(P<0.05).The immunofluorescence staining results showed that the expression of the raphe nuclei was lower in the model group than that in the normal group,the medication group and the electro-acupuncture group.The mRNA levels of P11,tPA and BDNF were lower in the model group than those in the normal group,the medication group and the electro-acupuncture group(P<0.01).Conclusion:Electro-acupuncture has obvious anti-depressant effect and can improve the behavior of rats with chronic stress depression.Electro-acupuncture can increase the mRNA content of P11,tPA and BDNF in the rat model of chronic stress depression.This anti-depressant effect may be through regulating P11-tPA-BDNF pathway.
