摘要
以粉彩的幼嫩茎段为外植体,通过不同培养基和激素的组配筛选出适宜粉彩愈伤形成、不定芽分化增殖和生根的最优培养基,以建立红山樱粉彩的最佳快繁体系。结果表明:诱导粉彩愈伤组织的培养基为1/4MS+6-BA1.0mg·L^-1+IBA0.02mg·L^-1,分化丛生芽的培养基为1/4MS+6-BA1.0mg·L^-1+IBA0.01mg·L^-1,分化率为43.33%,增殖的培养基为MS+6-BA0.5mg·L^-1+NAA0.1mg·L^-1+TDZ0.1~0.2mg·L^-1,最高增殖系数为503.33%,生根的培养基为3/4MS+NAA0.1mg·L-1+IBA0.1mg·L^-1,4d后陆续生根,生根率100%。移栽的基质比例为珍珠岩∶泥炭土∶蛭石=1∶2∶1,植株移栽成活率可达53.33%。通过茎段间接诱导丛生芽,可建立粉彩高效快繁技术体系。
In this paper,the young stem segments of Cerasus jamasakura variety Fencai were used as explants to study the optimal medium for Fencai callus formation,adventitious bud differentiation,proliferation and rooting,and finally established the best rapid propagation system of Fencai.The results showed that the medium for the induction of Fencai callus was 1/4 MS+6-BA 1.0 mg·L^-1+IBA 0.02 mg·L^-1,while the medium for fascicular buds differentiation was 1/4 MS+6-BA 1.0 mg·L^-1+IBA 0.01 mg·L^-1,with 43.33%differentiation rate.In addition,the medium for proliferation was MS+6-BA 0.5 mg·L^-1+NAA 0.1 mg·L^-1+TDZ 0.1-0.2 mg·L^-1,with 503.33%maximum proliferation coefficient.The rooting medium was 3/4 MS+NAA 0.1 mg·L^-1+IBA 0.1 mg·L^-1,and the rooting rate was 100%after 4 days.The matrix ratio of transplanting was perlite∶peat∶vermiculite=1∶2∶1,and the survival rate of plant transplanting was 53.33%.The technology system of high efficiency and fast propagation of pastel can be established by inducing fascicled buds indirectly through stem segments.
