低温环境下水稻种子活力QTL鉴定 被引量:7

摘要 【目的】提高水稻芽期低温抗性,发掘低温环境下水稻种子活力相关基因,可为培育抗冷品种提供新的基因和种质资源。【方法】以耐冷型粳稻龙稻5号和冷敏感型籼稻中优早8杂交衍生的重组自交系(recombinant inbred lines,RILs)群体为试验材料,鉴定自然和人工低温环境下种子活力相关的QTL。【结果】不同低温胁迫环境下,龙稻5号种子具有较强的活力,中优早8种子的发芽率均显著低于龙稻5号;群体中不同株系低温发芽率存在较大差异,籼粳属性影响水稻种子低温环境下发芽率,但相关性未达到显著水平。共检测到10个控制低温环境下种子活力相关的QTL,自然和人工低温胁迫环境下各检测到5个相关的QTL,分布于第2、3、7、8、11和12号染色体上,3个QTL在不同环境下共性表达;共检测到17对上位性互作位点,第1、2、3和12号染色体区域上位性效应明显,加性效应QTL qLTGN2和qLTG12参与上位性互作。【结论】自然和人工低温胁迫环境下种子活力表型和QTL有较好的重复性,稳定表达的QTL qLTG8、qLTG11和qLTG12对低温环境下发芽率具有明显的调控效应,且上位性互作也是调控水稻芽期耐冷性的重要组成部分。 [Objective]This study aimed to improve the low temperature resistance of rice in the germination stage,identify the stable expression of novel QTLs for the seed vigor under low temperature environment,which will provide more gene and germplasm resources for the cultivation of cold tolerance rice varieties.[Method]In this study,a RILs population derived from the cross between Longdao5 and Zhongyouzao8 was used to identify seed vigor by germination rate traits under natural and artificial low temperature environments.[Result]The germination rate of the parents had a significant difference under low temperature environment,the germination rate of Zhongyouzao8 was significantly lower than that of Longdao5,cold tolerance was affected by Fi but did not reash a significant level.A total of ten major QTL were detected,five related QTLs were detected in both the low temperature environments,these QTLs were distributed in chromosomes 2,3,7,8,11 and 12.Among these QTLs,qLTG8,qLTG11 and qLTG12 were stably detected under two environments.In addition,a total of 17 pairs of epistatic loci were detected,there was an obvious effect on the chromosomes 1,2,3 and 12,the major QTLs qLTGN2 and qLTG12 were involved in the epistatic interaction.[Conclusion]The phenotype and QTL of seed germination rate under natural and artificial low-temperature environments have good repeatability.Among these QTLs,qLTG8,qLTG11 and qLTG12 of the stable expression have an obvious effect on germination rate under low-temperature environment,and the epistatic interaction also plays an important part in the regulation of cold tolerance of rice at the germination stage.
