摘要
为了简化重组大肠杆菌生产番茄红素的工艺,该研究通过选择大肠杆菌中17个转录因子相关基因的启动子序列,以编码番茄红素合成基因的CrtEBI为目的基因,通过构建表达番茄红素合成基因的质粒,来评估转录因子相关基因启动子序列对番茄红素产量的影响。通过比较不同菌株合成番茄红素的能力,确定最佳启动子。结果表明,含有组成型启动子P lpp或P mglb的菌株合成番茄红素的能力较强,相较于传统诱导型启动子菌株产量分别提高了347.7%和51.3%。由于筛选到的启动子为组成型启动子,省去了添加诱导剂的过程,达到了优化生产工艺、降低生产成本的目的。
Lycopene is a kind of fat-soluble carotenoids with strong antioxidant properties.It is widely used in food,medicine and other industries.Previous studies have constructed engineered Escherichia coli with high-level production of lycopene using inducer.But these methods are labor intensive and time-consuming.In order to simplify the lycopene production,17 promoters that regulated by transcription factor in E.coli were selected and E.coli recombinants were constructed e by cloning promoter sequences and ligating with report gene crtEBI.E.coli recombinant with the promoter P lpp and P mglb were mediated more effective expression.The lycopene yield increased by 51.3%compared with the control strain pTRC99aEBI.
