黄芪多糖对肺气虚型变应性鼻炎大鼠TSLP、OX40L mRNA表达的影响 被引量:12

Effects of Astragalus Polysaccharide on mRNA Expression of TSLP,OX40L in Allergic Rhinitis Rats with Lung Qi Deficiency Syndrome

摘要 【目的】探讨黄芪多糖对肺气虚型变应性鼻炎大鼠鼻黏膜组织胸腺基质淋巴细胞生成素(TSLP)、OX40配体(OX40L)mRNA表达的影响。【方法】将40只SD大鼠分为正常组、模型组、氯雷他定组、黄芪多糖组,每组10只。除正常组外,其余3组均采用烟熏法结合卵清蛋白致敏法建立肺气虚型变应性鼻炎大鼠模型。在局部激发阶段,氯雷他定组给予氯雷他定灌胃,黄芪多糖组给予黄芪多糖灌胃,正常组和模型组灌胃等量生理盐水。给药结束后,观察肺气虚型证候表现,对变应性鼻炎行为学进行评分,计算大鼠脾脏指数,采用苏木素-伊红(HE)和天狼猩红染色观察鼻黏膜组织病理改变,采用酶联免疫吸附分析(ELISA)检测血清中白细胞介素4(IL-4)、干扰素γ(IFN-γ)水平,采用实时定量逆转录聚合酶链反应(RT-qPCR)法检测鼻黏膜组织TSLP、OX40L m RNA表达的水平。【结果】与正常组比较,模型组大鼠肺气虚证候表现明显,鼻炎症状评分升高,脾脏指数升高,鼻黏膜组织重塑病理表现明显,血清中IL-4水平升高、IFN-γ水平降低,鼻黏膜TSLP、OX40L mRNA表达量显著升高(均P <0.05)。与模型组比较,黄芪多糖组和氯雷他定组大鼠肺气虚证候表现明显改善,鼻炎症状评分降低,脾脏指数降低,鼻黏膜损伤减轻,血清中IL-4水平降低、IFN-γ水平升高,鼻黏膜TSLP、OX40L mRNA表达量明显降低(均P <0.05),且2个治疗组比较,差异无统计学意义(P> 0.05)。【结论】黄芪多糖可有效改善肺气虚型变应性鼻炎大鼠鼻黏膜炎性症状,其作用机制与下调鼻黏膜组织TSLP、OX40L mRNA表达有关。 Objective To explore the effects of astragalus polysaccharide on the mRNA expression of thymic stromal lymphopoietin(TSLP)and OX40 ligand(OX40 L)in nasal mucosa of allergic rhinitis rats with lung qi deficiency syndrome. Methods Forty SD rats were randomly divided into 4 groups,namely normal group,model group, loratadine group and astragalus polysaccharide group, 10 rats in each group. Apart from the normal group,the rats in the other 3 groups were induced into allergic rhinitis model with lung qi deficiency syndrome by using smoking method and sensitization with ovalbumin. During local stimulation modeling stage,the loratadine group was given intragastric administration of loratadine solution,and astragalus polysaccharide group was given intragastric administration of astragalus polysaccharide solution,and the normal group and model group were given intragastric administration of normal saline. After medication,the symptoms of lung qi deficiency were observed,the scoring of allergic rhinitis behaviors were performed,the rat spleen index was calculated,the pathological changes in nasal mucosa tissues were observed by haematoxylin andeosin(HE)staining and Sirius Red staining method, the serum levels of interleukin 4(IL-4)and interferon γ(IFN-γ)were detected by enzyme-linked immunosorbent assay(ELISA),the mRNA expression levels of TSLP and OX40 L in nasal mucosa were detected by real time quantitative polymerase chain reaction(RT-PCR). Results Compared with the normal group, the manifestation of lung qi deficiency syndrome was obvious in the model control group,rhinitis symptoms score of the rats was increased,spleen index was enhanced significantly,the pathological of manifestation of nasal mucosa tissue remodeling was seen,serum IL-4 level was increased,serum IFN-γ level was decreased,and the mRNA expression levels of TSLP and OX40 L in nasal mucosa were increased(all P < 0.05). Compared with the model group,the lung qi deficiency syndrome manifestation was improved in the astragalus polysaccharide group and loratadine group,nasal mucosal injury was relieved,rhinitis symptoms score of the rats was decreased,spleen index was increased significantly,serum IL-4 level was decreased,IFN-γ level was increased,and the mRNA expression levels of TSLP and OX40 L in nasal mucosa were increased(all P < 0.05),and there was no difference between the two treatment groups(P > 0.05). Conclusion Astragalus polysaccharide is effective for improving the nasal mucosal inflammatory symptoms in allergic rhinitis rats with lung qi deficiency syndrome,whose mechanism is related to down-regulating the mRNA expression of TSLP and OX40 L in nasal mucosa.
