摘要
本试验旨在研究黄芪多糖(APS)对脂多糖(LPS)刺激小鼠巨噬细胞形态及免疫功能的影响。采用LPS刺激小鼠巨噬细胞(RAW264.7细胞),研究不同浓度(0.1、0.5、1.0 mg/mL)的APS对LPS刺激引起的小鼠巨噬细胞免疫应激的调控作用。结果表明:1)1.0 mg/mL APS能抑制由LPS刺激引起的巨噬细胞形态和增殖能力的变化。2)1.0 mg/mL APS能够显著提高巨噬细胞酸性磷酸酶的活性(P<0.05)。0.1、0.5、1.0 mg/mL的APS能够显著缓解由LPS刺激引起的碱性磷酸酶活性的降低(P<0.05)。3)不同浓度APS均能显著降低由LPS诱导的促炎症因子白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)含量的升高(P<0.05)。4)LPS刺激显著提高巨噬细胞Toll样受体4(TLR4)、髓样分化因子88(MyD88)、核因子-κB(NF-κB)的mRNA表达量(P<0.05),而添加1.0 mg/mL APS后,巨噬细胞TLR4、MyD88、NF-κB的mRNA表达量均表现出显著降低(P<0.05)。综上可知,APS可以改善小鼠巨噬细胞形态,恢复细胞增殖能力,并可通过调节TLR4信号通路相关基因的表达降低促炎症因子IL-1β和TNF-α分泌,减少机体免疫应激反应。
The purpose of this study was to investigate the effects of Astragalus polysaccharides(APS)on the morphology and immune function of lipopolysaccharide(LPS)-stimulated macrophages in mice.LPS was used to stimulate mouse macrophages(RAW264.7 cells),and the effects of different concentrations(0.1,0.5 and 1.0 mg/mL)of APS on the regulation of cellular immune stress caused by LPS stimulation were studied.The results showed as follows:1)1.0 mg/mL APS can inhibit the changes of cell morphology and proliferation induced by LPS stimulation.2)1.0 mg/mL APS could significantly increase the activity of macrophage acid phosphatase(P<0.05),and 0.1,0.5 and 1.0 mg/mL APS could significantly alleviate the reduction in alkaline phosphatase activity induced by LPS stimulation(P<0.05).3)The increases of pro-inflammatory cytokines interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)contents induced by LPS were significantly decreased by different concentrations of APS(P<0.05).4)LPS stimulation significantly increased the mRNA expression levels of macrophages Toll-like receptor 4(TLR4),myeloid differential factor 88(MyD88)and nuclear factor-κB(NF-κB)(P<0.05),while the mRNA expression levels of macrophages TLR4,MyD88 and NF-κB significantly decreased after the addition of 1 mg/mL APS(P<0.05).In summary,APS can improve the macrophage morphology and restore cell proliferation ability of mice.By regulating the expression of genes related TLR4/NF-κB signaling pathway,the secretion of pro-inflammatory cytokines IL-1βand TNF-αare decreased to reduce the body’s immune stress response.
