电针对慢性氯胺酮给药大鼠海马细胞形态及Aβ1-42表达的影响

The effect of electroacupuncture on the morphology of cells and the expression of Aβ1-42 in hippocampus chronic ketamine administrated rats

摘要 目的:探讨慢性氯胺酮给药后大鼠海马CA1、CA3区细胞形态和β-amyloid 1-42(Aβ1-42)表达变化及电针对其表达的调节作用。方法:将24只SD健康雄性大鼠随机分为正常组(N组)、生理盐水组(S组)、氯胺酮组(K组)、氯胺酮+电针组(KE组),每组6只。K组经腹腔注射盐酸氯胺酮注射液,注射剂量为(50 mg·kg-1),每天一次,连续7 d。S组给予等量的生理盐水腹腔注射,每天一次,连续7 d。KE组与K组同时给药,KE组在每天给药后半小时取三阴交穴和足三里穴进行电针,停药后再继续电针7 d,共14 d。采用尼氏染色观察海马区的细胞形态,免疫荧光法检测各组大鼠海马CA1、CA3区Aβ1-42的表达情况。结果:尼氏染色结果显示K组海马CA1、CA3区尼氏染色显示部分神经细胞有空泡且排列不规则,KE组空泡的神经细胞数量较K组减少,细胞排列有所改善。免疫荧光染色显示K组海马CA1、CA3区Aβ1-42和荧光强度表达较N组和S组显著增加(P<0.01);KE组海马CA1、CA3区Aβ1-42和荧光强度表达表达较K组显著降低(P<0.01)。结论:电针治疗可改善慢性氯胺酮给药大鼠海马的神经细胞异常变化,并下调氯胺酮诱导的海马CA1、CA3区Aβ1-42的过表达。 Objective:To investigate the changes of cell morghology and the expression of β-amyloid 1-42(Aβ1-42) in hippocampus CA1 and CA3 of rats after chronic ketamine administration, and the regulatory effect of electroacupuncture. Methods:Twenty-four SD male rats were randomly divided into 4 groups(n=6/group): normal group(N), saline group(S), ketamine group(K)and ketamine + electroacupuncture group(KE).The Group K and Group KE were intraperitoneally with ketamine(50 mg·kg-1) once a day for 7 days. The Group S were given normal saline intraperitoneally once a day for 7 days. The Group KE were given electroacupuncture at Sanyinjiao and Zusanli points half an hour after intraperitoneally with ketamine and once daily for 7 days after ketamine withdrawal,14 days in total. The morphology of hippocampus was observed by Nissl staining. The expression of Aβ1-42 in hippocampus CA1 and CA3 of each group was detected by immunofluorescence. Results:Nissl staining showed that there were some vacuoles and irregular arrangement in CA1 and CA3 area of hippocampus in Group K. Compared with the Group K, the number of vacuoles in Group KE was less and the arrangement of cells were improved. Immunofluorescence staining showed that compared with the Group N and S the expression of Aβ1-42 and fluorescence intensity in CA1 and CA3 area of hippocampus in Group K were significantly higher(P<0.01). Compared with the Group K, the expression of Aβ1-42 and fluorescence intensity in CA1 and CA3 area of hippocampus in Group KE were significantly lower(P<0.01). Conclusion:Electroacupuncture can improve the abnormal changes of neurons in hippocampus of chronic ketamine-administered rats and down-regulate the overexpression of Aβ1-42 in hippocampal CA1 and CA3 induced by ketamine.
