摘要
目的研究莲心碱对七氟烷诱发新生大鼠皮层癫痫样脑电图波的影响及机制。方法将SD新生大鼠随机分为模型组(n=15)、对照组(n=15)和低(n=15)、高(n=15)剂量实验组。将各组大鼠放入麻醉箱中吸入6.0%七氟烷3 min进行麻醉诱导,然后持续吸入2.1%七氟烷维持麻醉,持续1 h。低、高剂量实验组于麻醉后30 min腹腔注射2.5、5.0g·L^-1莲心碱10μL,对照组给予100 g·L^-1左乙拉西坦10μL,模型组腹腔注射生理盐水5μL·g^-1。以血气分析仪检测各组大鼠血气指标,以酶联免疫吸附(ELISA)法检测血清皮质醇含量,监测大鼠皮层脑电图,以蛋白质印迹法检测各组大鼠海马组织中pAkt、pS6、pAMPK蛋白的表达。结果模型组、对照组及低、高剂量实验组大鼠癫痫波总时长分别为(45.56±9.23),(13.56±5.41),(37.53±7.53),(12.36±5.69)s,平均时长分别为(8.56±3.35),(2.69±0.33)(6.69±2.52),(2.36±0.25)s,癫痫波个数分别为(7.25±1.24),(1.52±0.42),(5.16±0.56),(1.69±0.58)个;血清中皮质酮含量分别为(416.28±23.58),(124.05±62.18),(362.58±46.28),(139.52±37.85)ng·mL^-1,差异均有统计学意义(P<0.05)。结论莲心碱可剂量依赖性抑制七氟烷诱发的新生大鼠皮层癫痫样脑电图波,可能通过调节mTOR通路进而发挥脑保护作用的。
Objective To study the effect and mechanism of liensinine on the epileptic-like electroencephalo-graph(EEG)in the cortex of neonatal rats induced by sevoflurane.Methods SD eonate rats were randomly divided into Exp-L/H groups(n=15),model group(n=15)and control group(n=15).Each group of rats was put into an anesthesia box for inhalation of 6.0%sevoflurane for 3 min for anesthesia induction,followed by continuous inhalation of 2.1%sevoflurane for 1 h.Exp-L/H groups were injected with 2.5,5.0 g·L^-1 liensinine 10μL in 30 min after anesthesia,and 100 g·L^-1 letiracetam 10μL was administered to control group.Model group was intraperitoneally injected with 5μL·g^-1 normal saline.Blood gas of each group was detected by blood gas analyzer.The content of serum cortisol was detected by ELISA,and the expression of pAkt,pS6 and pAMPK in the hippocampus of each group was detected by Westen blotting.Results The total length of the epileptic waves in model group,control group and Exp-L/H groups were(45.56±9.23),(13.56±5.41),(37.53±7.53),(12.36±5.69)s,the average duration were(8.56±3.35),(2.69±0.33),(6.69±2.52),(2.36±0.25)s,the number of epileptic waves were 7.25±1.24,1.52±0.42,5.16±0.56,1.69±0.58,the content of corticosterone in the serum were(416.28±23.58),(124.05±62.18),(362.58±46.28),(139.52±37.85)ng·mL^-1,all with significant difference(all P<0.05).Conclusion Liensinine can inhibit epileptic electroencephalogram(EEG)waves induced by sevoflurane in neonatal rats in a dose-dependent manner,which may play a protective role by regulating mT OR pathway.
