枯草芽孢杆菌XL05菌株的紫外诱变及发酵配方优化 被引量:4

摘要 枯草芽孢杆菌(Bacillus subtilis)XL05是从土壤中筛选的一株拮抗细菌,该菌株抗菌谱广,对根腐病原菌、绿僵菌和烟草赤星病菌等多种植物病原真菌具有强烈抗性。为获得高产抗真菌活性物质的突变菌株,对筛选到的原始菌株进行紫外诱变,并确定其遗传稳定性;选择遗传稳定性最好的突变株进行发酵配方优化。试验结果表明:该菌株的紫外诱变的最佳照射时间为20 s,通过筛选,获得5株较稳定的高产突变株,其效价较出发菌株均提高1倍以上,且遗传性状稳定;ZW75突变株最优配方为葡萄糖4.0%、蛋白胨0.2%和CaCO 30.1%,为该突变株的工业发酵提供一定的理论基础。 Bacillus subtilis XL05 was an antagonistic strain isolated from soil.The strain has a wide antibacterial spectrum and strong resistance to root rot pathogen,Metarhizium,Alternaria alternata and other plant pathogenic fungi.In order to obtain mutant strains with high yield of fungal active substances,the selected wild strains were mutated by UV and their genetic stability was determined.The mutant strain with the best genetic stability was selected for optimization on fermentation medium formula.The results showed that the optimal irradiation time of UV mutagenesis of this strain was 20 s.Through screening,5 stable mutant strains with high yield were obtained.Tantagonistic potency of each strainwas more than 1 time higher than that of the original strain and their genetic characteristics were stable.The optimal formula of ZW75 mutant was glucose 4.0%,peptone 0.2%and calcium carbonate 0.1%,which provided a theoretical basis for the industrial fermentation by the mutant.
