摘要
目的:观察阳明经合穴刺络放血法对特应性皮炎(atopic dermatitis,AD)模型小鼠病理学及血清IgE水平的影响。方法:将24只C57bl/6J小鼠随机平均分为阳明经合穴刺络放血组、皮损局部刺络放血组、模型组、空白组。除空白组外,阳明经合穴刺络放血组、皮损局部刺络放血组、模型组通过半抗原诱导法造建立AD模型。阳明经合穴刺络放血组取手足阳明经合穴,即曲池、足三里刺络放血,皮损局部刺络放血组在皮损周边局部刺络放血,模型组、空白组只抓取,不治疗。造模与治疗交替进行,共治疗8次。16 d后,记录各组皮损评分,取皮损处皮肤HE染色镜下观察,酶联免疫吸附法测定血清IgE水平。结果:与空白组比较,模型组皮损评分及IgE水平显著上升(P<0.01,P<0.01);与模型组比较,皮损局部刺络放血组皮损评分明显下降(P<0.05),阳明经合穴刺络放血组皮损评分及IgE水平明显下降(P<0.05);皮损局部刺络放血组与阳明经合穴刺络放血组造模处皮损评分及IgE水平比较,差异无统计学意义(P>0.05)。病理学观察显示,与正常皮肤比较,模型组皮损处表皮和真皮皮肤明显增厚,大量炎症细胞浸润,并伴有表皮角化不全。阳明经合穴刺络放血组、皮损局部刺络放血组小鼠皮肤增厚及角化不全不及模型组,有中等量炎细胞浸润。阳明经合穴刺络放血组、皮损局部刺络放血组差异不明显。结论:阳明经合穴刺血法可以减轻AD模型小鼠皮损程度,这可能与阳明经合穴刺络放血法能下调小鼠血清IgE水平有关。
Objective:To observe the effect of pricking blood at Yangming Meridian Meridian Points on the pathology and serum IgE level of atopic dermatitis(AD)model mice.Methods:Twenty-four C57bl/6J mice were randomly divided into a bloodletting group at Yangming Meridian Points,a bloodletting group with local skin lesions,a model group,and a blank group.In the Yangming Meridian Meridian puncture and bloodletting group and the skin lesion local puncture and bloodletting model group,the atopic dermatitis model was established by the hapten induction method,and the blank group was not modeled.In the Yangming Meridian and bloodletting group,take the Yangming Meridian points of the hands and feet,namely Quchi and Zusanli for bloodletting.In the local puncture and bloodletting group of the skin lesions,the bloodletting was performed around the skin lesions.,No treatment.Modeling and treatment were performed alternately,with a total of 8 treatments.16 days later,the scores of the skin lesions in each group were recorded,and the skin lesions were taken and observed under a microscope with HE staining.The serum IgE level was determined by enzyme-linked immunosorbent method.Results:Compared with the blank group,the skin lesion score and IgE level of the model group increased significantly(P<0.01,P<0.01);compared with the model group,the skin lesion score of the local puncture bloodletting group significantly decreased(P<0.05).The scores of skin lesions and IgE levels in the bloodletting group at the Yangming Meridian Meridian Meridian Points were significantly decreased(P<0.05);the skin lesion scores of the modelling sites in the local piercing and bleeding group and the Yangming Meridian Meridian Meridian bleeding group Compared with IgE levels,the difference was not statistically significant(P>0.05,P>0.05).Pathological observation showed that compared with normal skin,the epidermis and dermal skin of the skin lesions in the model group were significantly thickened,and a large number of inflammatory cells were infiltrated,accompanied by epidermal insufficiency.The skin thickening and incomplete keratosis of mice in the bloodletting group of Yangming Meridian Points and the bloodletting group of local skin lesions were less than those of the model group,with moderate inflammatory cell infiltration.There was no significant difference between the bloodletting group at Yangming Meridian Points and the bloodletting group with local skin lesions.Conclusion:The method of pricking blood at the combined points of Yangming Meridian can alleviate the degree of skin lesions in mice with atopic dermatitis,which may be related to that the method of pricking blood at the combined points of Yangming Meridian can down-regulate the level of serum IgE in mice.
