摘要
利用纤维素酶协同超声波提取技术对鱼腥草中总黄酮的提取工艺进行筛选,同时考察黄酮对DPPH自由基的清除作用。以芦丁为对照品,黄酮提取率为指标,通过单因素试验和L9(3^4)正交试验优选出纤维素酶协同超声波法提取总黄酮的最佳工艺为:乙醇浓度60%、超声时间30min和超声功率600w,即A2B3C2,其因素影响力大小为:B>A>C。在该提取条件下黄酮得率为2.8227%。鱼腥草中黄酮类化合物在一定范围内对DPPH自由基有显著的清除作用,且随着黄酮类化合物浓度的增加清除作用增强。本实验优选的工艺设计合理,稳定可行,抗氧化活性较好,为鱼腥草的进一步研究提供技术基础。
The extraction process of total flavonoids from Houttuynia cordata was screened by using cellulase synergistic ultrasonic extraction technology.At the same time,the clear effect of flavonoid extract on DPPH free radicals was investigated.With rutin as the reference substance,the extraction rate of flavonoids was the index.The optimum conditions for the extraction of total flavonoids by cellulase and ultrasonic method were optimized by single factor test and L9(34)orthogonal test.The ultrasonic treatment time is 30min,ethanol volume fraction is 60%,ultrasonic treatment power is 600w,ie A2B3C2,the factor influence size is:B>A>C.The extraction rate of flavonoids under this process condition was 2.8277%.The flavonoids from Herba Houttuyniae have a clear effect on DPPH free radicals,and in a certain range,the clear action of flavonoids increases with the increase of flavonoids content.The optimized extraction process was scientific,stable and feasible,can provided a new basis for the comprehensive assessment and overall control of the quality of Houttuynia cordata Thunb.
