摘要
为探究2018年7月广西南宁市武鸣区某规模化肉猪场保育猪急性死亡的病因,本研究通过细菌的分离鉴定、16S rRNA的扩增、生物被膜体外形成条件的测定、毒力基因检测、药物敏感性试验、耐药基因检测等试验方法,对可能存在的病原菌的生物学特性及耐药性进行分析。结果表明:该菌为稍直或弯曲的革兰氏阳性杆菌,且与猪红斑丹毒丝菌在基因上的同源性最高,鉴定为猪红斑丹毒丝菌。该菌株生长迅速,在室温28℃时具有较强的生物被膜体外形成能力;毒力基因检测结果表明含有荚膜多糖CPS-A、黏附素RspA、表面保护性抗原SPaA共3种毒力基因;该菌株耐药性强,耐药率高达68.18%,仅对米诺环素、多西环素、恩诺沙星、氟苯尼考4种药物敏感,并扩增出blaTEM(788 bp)、Sul2(435 bp)两种耐药基因,其耐药表型与耐药基因检出呈现正相关。本研究将为预防猪红斑丹毒丝菌的传播及临床用药提供指导性意见。
To explore the cause of acute death of pigs in July 2018 on a large-scale pig farm in Wuming district,Nanning,Guangxi province,this study analyzed the biological characteristics and drug resistance of the causative agents through isolation and identification of bacteria,amplification of 16 sRNA,determination of conditions for formation of biofilm in vitro,drug sensitivity test,detection of drug resistance genes,etc.One bacterial isolate was obtained,identified and characterized to investigate its relation to the acute death.This strain was a slightly straight or curved Gram-positive bacterium and had the highest genetic homology with Erysipelothrix rhusiopathiae.The strain grew rapidly and had strong biofilm formation ability at room temperature of 28℃.In addition,this strain was highly drug resistant.It was susceptible to minocycline,doxycycline,enrofloxacin and florfenicol only.Two drug resistance genes blaTEM(788 bp)and Sul2(435 bp)were amplified and related to the drug resistant phenotype.This study would provide guidance for the prevention of the transmission and clinical treatment of E.rhusiopathiae.
