摘要
Hedgehog(HH)信号通路是促进胚胎分化、系统发育和组织调控的重要信号传导系统,突变、非经典转录调控及表观遗传变化可引起该通路的异常激活。已证实,该通路异常激活可导致发育障碍及各种恶性肿瘤。目前认为,该通路对发育和某些恶性肿瘤(如基底细胞癌、髓母细胞瘤及横纹肌肉瘤等)的发生及进展发挥着不可替代的作用。鉴于HH通路成员的表观遗传异常在恶性肿瘤发病机制的特殊性及其可作为某些恶性肿瘤(如白血病、胃癌)潜在的治疗靶点,其已成为HH通路的研究热点。为对恶性肿瘤发病机制及潜在治疗靶点提供新的认识,本文综述了HH信号通路成员的表观遗传异常及其对恶性肿瘤发生调控方面的研究进展。
The Hedgehog(HH) signaling pathway is a vital signaling system that promotes embryonic differentiation, phylogeny, and tissue regulation. Mutations, non-canonical transcriptional regulation,and epigenetic changes can cause aberrant activation of this pathway. It has been confirmed that abnormal activation of this pathway can lead to developmental disorders and various malignant tumors. This pathway plays an irreplaceable role in the development,occurrence and progression of certain malignant tumors,such as basal cell carcinoma,medulloblastoma,and rhabdomyosarcoma. Considering the particularity of the mechanism of the epigenetic abnormality of HH pathway members,and the potential for therapeutic targets,in certain malignant tumors,such as leukemia and gastric cancer,the HH pathway has become an important research topic. In order to provide a new understanding of the pathogenesis of malignant tumors and potential therapeutic targets,this article reviews the epigenetic abnormalities of members of the HH signaling pathway and the research progress in the regulation of malignant tumors.
