摘要
目的基于RAW264.7细胞模型探讨荷叶提取物的抗炎活性并初步探讨其作用机制,为中药药效物质基础与作用机制提供理论依据。方法采用溶剂提取法、水提醇沉法等将荷叶水提物(NNE)分为荷叶多糖部位(NNP)、荷叶正丁醇部位(NNB)和荷叶氯仿部位(NNC);采用MTT法筛选活性部位并确定药物的给药质量浓度;Elisa法检测荷叶各部位对由脂多糖(LPS)诱导的细胞炎症因子肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)和NO表达水平的影响。结果NNE、NNP和NNB均能降低细胞炎症模型胞外炎症介质TNF-α、IL-6和NO的释放。结论荷叶提取物具有抗炎活性,为荷叶的进一步开发利用提供了理论依据。
Objective To investigate the anti-inflammatory activity of leaves of Nelumbo nuxifera Gaerth extract based on RAW264.7 cell model,and to discuss the mechanism for providing a theoretical basis for development and utilization of leaves of Nelumbo nuxifera Gaerth.Methods Leaves of Nelumbo nuxifera water extract(NNE)was fractionated into Nelumbo nuxifera polysaccharide fraction(NNP),Nelumbo nuxifera n-butanol fraction(NNB)and Nelumbo nuxifera chloroform fraction(NNC)by solvent extraction method and ethanol subsiding method.The active sites and the appropriate drug concentration were determined by MTT.The effect of different fraction of leaves of on the expression of lipopolysaccharide(LPS)-induced cytokines level tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and NO was detected by Elisa.Results NNE,NNP and NNB reduced the release of TNF-α,IL-6 and NO in different degrees.Conclusion Leaves of Nelumbo nuxifera Gaerth extract have the anti-inflammatory activity and provides theoretical foundation for further development and utilization of leaves of Nelumbo nuxifera Gaerth.
