姜黄素对心肌缺血再灌注大鼠心肌组织保护作用的研究 被引量:2

摘要 目的:研究姜黄素对心肌缺血再灌注大鼠心肌组织的保护作用。方法:采用结扎左冠状动脉前降支法建立心肌缺血再灌注模型,随机将模型大鼠分为模型组、卡托普利组(60 mg·kg^-1)、姜黄素400 mg·kg^-1组、姜黄素200 mg·kg^-1组,每组10只。各组予相对应药物连续灌胃给药12周,1次/d。另设正常组大鼠10只。正常组、模型组大鼠灌胃予等体积的生理盐水。给药12周后,采用MS4000U-1C型生物信号定量记录分析系统检测大鼠心脏动力学指标平均动脉血压(MAP)、心率(HR)、左心室舒张末期压(LVEDP)的水平。苏木精-伊红(HE)染色观察心肌组织的病理学变化。ELISA法检测血清中心肌酶肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)的含量。试剂盒检测心肌组织中超氧化物歧化酶(SOD)的活性和丙二醛(MDA)的含量。ELISA法检测心肌组织中炎症因子白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的含量。Western blotting检测心肌组织中TLR4、Bcl-2、Bax蛋白的表达。结果:与模型组比较,姜黄素各剂量组心肌病理学变化得到改善,MAP、HR显著升高,LVEDP明显降低,血清及心肌组织中CK、CK-MB、LDH、IL-1β、IL-6、TNF-α含量明显降低,心肌组织中SOD活性增加,MDA含量减少,TLR4、Bax蛋白表达减少,Bcl-2蛋白表达增加(均P<0.05)。结论:姜黄素对心肌缺血再灌注大鼠的心肌组织具有保护作用,其作用机制可能与减少心肌酶释放以及提高机体组织抗氧化能力有关。 Objective:To study the protective effect of curcumin on myocardial tissues of rats with myocardial ischemia/reperfusion injury.Methods:A rat model of myocardial ischemia/reperfusion was established by ligation of the anterior descending left coronary artery,and the rats were randomly divided into a model group,a captopril group(60 mg·kg-1),a curcumin 400 mg·kg-1 group and a curcumin 200 mg·kg-1 group.Each group was given the corresponding drug by gavage for 12 weeks,1 time per day.A normal group of 10 rats was also set up.Equal volume of saline was administered by gavage to rats in the normal and model groups.After 12 weeks of drug administration,mean arterial pressure(MAP),heart rate(HR)and left ventricular end-diastolic pressure(LVEDP)were measured by MS4000U-1C quantitative bio-signal recording and analysis system.The pathological changes of myocardial tissues were observed by hematoxylin-eosin(HE)staining.ELISA was used to determine the contents of creatine kinase(CK),creatine kinase isoenzyme MB(CK-MB)and lactate dehydrogenase(LDH)in serum.The contents of the inflammatory factors interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in myocardial tissues was also measured by ELISA.The activity of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)in myocardial tissues was detected using assay kits.Western blotting was used to determine the expression of TLR4,Bcl-2 and Bax protein in myocardial tissues.Results:Compared with the model group,myocardial pathological changes were improved in both curcumin groups,whereas MAP and HR increased significantly,LVEDP decreased significantly,CK,CK-MB,LDH,IL-1β,IL-6 and TNF-αcontents in serum and myocardial tissues decreased significantly,SOD activity increased,MDA content decreased,and TLR4 and Bax protein expression in myocardial tissues decreased(all P<0.05).Conclusion:Curcumin has a protective effect on myocardial tissues of myocardial ischemia/reperfusion rats and its mechanism may be related to the reduction of myocardial enzyme release and the enhancement of antioxidant capacity.
