摘要
为探讨柞蚕雄蛾粉对小鼠结肠炎的治疗效果及作用机制,采用自由饮用3%硫酸葡聚糖钠盐(DSS)的方法建立小鼠慢性结肠炎模型,连续7 d灌胃不同剂量(0.5 g/kg、2.5 g/kg、5 g/kg)的柞蚕雄蛾粉,调查灌胃柞蚕雄蛾粉后小鼠疾病活动指数(disease activity index,DAI)、血清免疫球蛋白G(IgG)含量和肠粘膜分泌型免疫球蛋白A(SIgA)含量,以及结肠炎性细胞因子IL-6、IL-4和TNF-α编码基因mRNA表达水平的变化。结果显示,与空白对照组小鼠相比,DSS诱导组小鼠的体质量、结肠长度、结肠质量、血清IgG含量、结肠粘膜SIgA含量、结肠IL-4 mRNA表达水平显著降低,DAI评分及结肠IL-6、TNF-αmRNA表达水平显著升高,结肠粘膜严重破坏,腺体萎缩,全层炎性细胞浸润;与灌胃羧甲基纤维素钠的DSS诱导组小鼠相比较,灌胃柞蚕雄蛾粉的各试验组小鼠体质量、结肠长度、结肠质量、血清IgG含量、结肠粘膜SIgA含量、结肠IL-4 mRNA表达水平显著升高,DAI评分及结肠IL-6、TNF-αmRNA表达水平显著降低,结肠组织炎性细胞减少,腺体萎缩不明显,黏膜结构较为完整。研究结果表明,柞蚕雄蛾粉对DSS诱导所致小鼠慢性结肠炎有一定的改善作用,其作用机制可能与调节炎性细胞因子的表达有关。
In order to explore the therapeutic effect and functional mechanism of male tussah moth powder on colitis in mice,a mouse chronic colitis model was established by free drinking of 3%dextran sulfate sodium salt(DSS).After different doses of male tussah moth powder(0.5 g/kg,2.5 g/kg,5 g/kg)were given to mice for 7 consecutive days by intragastric administration,disease activity index(DAI),changes in immunoglobulin G(IgG)content and intestinal mucosal secretory immunoglobulin A(SIgA)content as well as changes in the mRNA expression levels of the genes encoding the inflammatory cytokines IL-6,IL-4,and TNF-αwere investigated.The results showed that compared with the mice in blank control group,body weight,colon length,colon weight,IgG content in serum,SIgA content in intestinal mucosa,colon mRNA expression level of IL-4 in DSSinduced mice were significantly reduced,DAI score and mRNA expression levels of colon IL-6 and TNF-αwere significantly increased,colon mucosa was severely damaged with gland atrophy and full-thick inflammatory cell infiltration.Compared with DSS-induced mice fed with sodium carboxymethyl cellulose(CMC-Na),body weight,colon length,colon weight,IgG content in serum,SIgA content in intestinal mucosa,and mRNA expression level of colon IL-4 in mice fed with male tussah moth powder were significantly increased.DAI score and mRNA expression levels of colon IL-6 and TNF-αwere significantly reduced,colonic inflammatory cells were reduced,gland atrophy was not obvious,and the mucosal structure was relatively complete.The results show that the male tussah moth powder can improve DSS-induced chronic colitis in mice,and its functional mechanism may be related to the regulation of the expression of inflammatory cytokines.
