摘要
目的:应用网络药理学研究四物汤治疗早发性卵巢功能不全的靶点及作用机制。方法:通过TCMSP数据库获取四物汤的活性成分,利用TCMSP、Drug Bank、Swiss Target Prediction获得活性成分和激素补充治疗对应的靶点,利用Gene Cards获得早发性卵巢功能不全的相关靶点,取共有靶点使用STRIN、Cytoscape构建PPI网络图,并利用David对所获靶点进行通路富集分析。结果:四物汤中有20种活性成分对应217个靶点,早发性卵巢功能不全共有2724个靶点,激素补充治疗共有283个靶点,四物汤与早发性卵巢功能不全共有靶点138个,激素补充治疗与早发性卵巢功能不全共有靶点167个,三者共有靶点54个。GO富集显示四物汤与早发性卵巢功能不全共有靶点参与类固醇激素受体活性、雌二醇反应、类固醇激素介导通路等多项生物过程。KEGG富集显示共有靶点在卵巢类固醇合成、类固醇生物激素合成等受体信号通路上有明显富集。结论:四物汤是通过多靶点、多通路治疗早发性卵巢功能不全,与激素补充治疗有部分类似的机制。
Objective: To apply network pharmacology to predict the target and mechanism of Siwu Decoction(SWD) in the treatment of primary ovarian insufficiency(POI). Methods: Active ingredients of SWD were obtained through TCMSP database, target corresponding to active ingredients and hormone replacement therapy(HRT) were obtained by TCMSP, Drug Bank, Swiss Target Prediction database, POI related targets were obtained by genecard database, PPI network graph of common targets was constructed by using STRING database and Cytoscape software, and pathway enrichment analysis of target was conducted by using David database. Results:20 kinds of active ingredients in SWD corresponds to 217 targets, POI has a total of 2724 targets, HRT has a total of 283 targets, SWD and POI share targets for 138, HRT and POI for 167, a total of three targets for 54.GO enrichment analysis showed SWD and POI of targets of steroid hormone receptor activity, response to estradiol, and so on many biological processes of steroid hormones mediated pathway, KEGG concentration showed that there are targets in ovarian steroid synthesis, steroid hormone synthesis receptors signaling pathways, having obvious enrichment on the signal path. Conclusion: SWD treats POI through multi-target and multi-pathway therapy, which is partly similar to HRT.
