青钱柳对阴虚型2型糖尿病大鼠糖脂代谢的影响 被引量:2

摘要 目的探讨青钱柳对阴虚型2型糖尿病大鼠证侯指标及糖脂代谢的影响。方法雄性SD大鼠60只,将大鼠随机分为空白组和造模组。其中空白组7只给予普通饲料,造模组53只给予高脂饲料喂养8周。第10周,以30 mg/kg的剂量一次性腹腔注射2%链脲佐菌素(streptozotocin,STZ),并于72小时后选择空腹血糖>11.1 mmol/L的成模大鼠随机分为模型组,知柏地黄丸组,青钱柳水提物(cyclocarya paliurus water extract,CPE)低、中、高剂量组;空白组和模型组给予0.9%氯化钠溶液,知柏地黄丸组给予知柏地黄丸混悬液(0.3 g/kg),CPE低、中、高剂量组分别给予青钱柳水提取物[CPE(0.25、0.5、1 g/kg)]。连续给药4周,给药期间检测空腹血糖(fasting blood glucose,FBG)。给药4周后,检测三碘甲状腺原氨酸(triiodothyronine,T3)、甲状腺素(thyroxine,T4)、环磷酸腺苷(cyclic adenosine monophosphate,cAMP)、环磷酸鸟苷(cyclic guanosine monophosphate,cGMP)等含量。结果与模型组比:(1)不同剂量CPE组大鼠FBG明显降低,血糖曲线下面积(area under curve,AUC)明显降低,TC、TG、LDL水平明显降低(P<0.01,P<0.05);(2)不同剂量CPE组cAMP、cGMP、T3、T4水平明显降低(P<0.01,P<0.05);(3)不同剂量CPE组游离胰岛素(Free insulin,FINS)水平和胰岛素抵抗指数(Homeostasis model assessment for insulin resistance,HOMA-IR)明显降低(P<0.01,P<0.05)。结论青钱柳可缓解阴虚型T2D大鼠胰岛素抵抗和糖耐量损伤、调节血脂代谢紊乱的同时,还可改善T3、T4、cAMP、cGMP等证候指标。 Objective To investigate the effects of Cyclocarya paliurus on syndrome indicators and glycolipid metabolism of type 2 diabetes mellieus(T2D)rats with yin-deficiency type.Methods 60 male SD rats were randomly divided into blank group and modeling group.Among them,7 rats in blank groups were given ordinary feed only,53 rats in modeling group were fed with high-fat diet for 8 weeks only.At the 10th week,The modeling group was intraperitoneally injected 2%streptozotocin(STZ)at a dose of 30 mg/kg.After 72 hours,the rats in modeling group with fasting blood glucose higher than 11.1 mmol/L were randomly divided into model group,Zhibai Dihuang pills group,low,medium and high dose of Cyclocarya paliurus water extract(CPE)group.The blank group and model group were given 0.9%sodium chloride solution,Zhibai Dihuang pills group was given suspension of Zhibai Dihuang pills(0.3 g/kg),the low,medium and high dose of CPE group were given CPE with the concentrations of 0.25 g/kg,0.5 g/kg and 1 g/kg,respectively.Continuous administration for 4 weeks,the fasting blood glucose(FBG)was detected during the administration.After 4 weeks,the contents of triiodothyronine(T3),thyroxine(T40),cyclic adenosine monophosphate(cAMP)and cyclic guanosine monophosphate(cGMP)were detected.Results Compared with model group:(1)the serum FBG level of the rats in different doses of CPE groups decreased significantly,and the area under the curve(AUC)of blood glucose decreased significantly,the levels of TC,TG and LDL in serum all decreased significantly(P<0.05 or P<0.01);(2)The levels of cAMP,cGMP,T3 and T4 in different doses of CPE groups all decreased significantly(P<0.01 or P<0.05);(3)The level of free insulin(FINS)and homeostasis model assessment for insulin resistance(HOMA-IR)decreased significantly in different doses of CPE groups(P<0.05 or P<0.01).Conclusion Cyclocarya paliurus can alleviate the insulin resistance and impaired glucose tolerance of T2D rats with yin-deficiency type,regulating lipid disorders,while improving T3,T4,cAMP,cGMP and other syndrome indicators.
