钙敏感受体参与镉致原代大鼠成骨细胞损伤 被引量:1

Calcium sensing receptor is involved in Cd-induced cellular injury in primary rat osteoblasts

摘要 为探究钙敏感受体(CaSR)在镉致原代大鼠成骨细胞损伤中的作用,本研究用不同浓度的镉(1、2、5和10μmol/L)处理原代大鼠成骨细胞3 h,或用2μmol/L镉处理成骨细胞不同时间(10 min、30 min、1 h、3 h、6 h和12 h),或用2μmol/L镉与CaSR抑制剂/激活剂共处理成骨细胞3 h。通过Western-blot检测CaSR蛋白表达水平,q RT-PCR检测CaSR基因转录水平,Hoechst33258染色观察细胞核形态变化。结果显示,不同浓度的镉处理以及2μmol/L镉处理成骨细胞不同时间,均能显著增强成骨细胞中CaSR蛋白的表达水平;镉处理显著升高成骨细胞中CaSR基因的转录水平;CaSR抑制剂可缓解镉对成骨细胞细胞核的损伤,CaSR激活剂加剧镉对成骨细胞细胞核的损伤。上述结果表明,Ca SR在镉致原代大鼠成骨细胞损伤过程中发挥着重要作用。 In this study,in order to explore the role of Calcium sensing receptor(Ca SR)in Cadmium(Cd)induced-cellular injury in primary rat osteoblasts,osteoblasts were treated with different concentrations(1,2,5 and 10μmol/L)of Cd for 3 h or 2μmol/L Cd for different times(10 min,30 min,1 h,3 h,6 h and 12 h)or co-treat 2μmol/L Cd with Ca SR inhibitor/activator for 3 h.Western-blot was used to detect the expression level of Ca SR,q RT-PCR was used to detect the transcription level of Ca SR,Hoechst 33258 staining was used to observe the morphology changes in cell nuclear.The results showed that different concentrations of Cd and 2μmol/L Cd treatment for different times significantly enhanced Ca SR expression level in osteoblasts.Cd significantly increased Ca SR transcription level in osteoblasts.Ca SR inhibitor efficiently alleviated nuclear injury induced by Cd,Ca SR activator aggravated nuclear injury induced by Cd.Taken together,Ca SR plays an important role in Cd induced cellular injury in primary rat osteoblasts.
