摘要
使用超临界CO2萃取法提取黑麦籽粒多酚,以黑麦籽粒多酚得率为测定指标,通过单因素实验、正交实验设计结合响应面法,分析夹带剂种类、料液比、提取时间、压强、温度对黑麦籽粒多酚得率的影响并优化提取工艺,并运用高效液相色谱-质谱(HPLC-MS)对黑麦籽粒多酚提取液中主要成分进行结构鉴定。结果表明:超临界CO2萃取黑麦籽粒多酚工艺的最优条件为:夹带剂95%乙醇,料液比1:4 g/mL,萃取压强5230 psi,萃取时间2.07 h,萃取温度55.65℃,此条件下黑麦籽粒多酚得率为0.201 mg/g。初步分析表明,黑麦籽粒多酚中含有罗汉松脂酚、二十三烷基间苯二酚、二十五烷基间苯二酚和β-谷甾醇油酸酯。
Polyphenols from rye grain were extracted by supercritical fluid extraction-CO2 method.Single-factor experiment,orthogonal design and response surface methodology were used to analyze the effects of types of cosolvent,solid-liquid ratio,extraction time,pressure and temperature on the extraction rate of polyphenols from rye grains as a measurement index,and to optimize the extraction process.The components of polyphenols from rye grain extract were identified by means of high-performance liquid chromatography-mass spectrometry(HPLC-MS).The results showed that optimum process parameters extracting rye grain polyphenols by supercritical CO2 extraction method were as followed:95%ethanol as cosolvent,solid-liquid ratio of 1∶4 g/mL,extracted for 2.07 h at 55.65℃and 5230 psi,and the yield of rye grain polyphenols was 0.201 mg/g.Four polyphenols from rye grain were preliminarily identified as matairesinol,tricosylresorcinol,pentacosenylresorcinol and sitosteryl-18∶1.
