摘要
目的探讨胸腺肽α1对哮喘大鼠IL-25表达的影响,以及对Th2/Th1平衡的调节作用及相关机制,为哮喘的临床治疗提供实验依据。方法雄性SPF级Wistar雄性大鼠随机分为5组:正常组、哮喘模型组、地塞米松治疗组(0.5 g/kg)、胸腺肽α1治疗组(5 mg/kg),地塞米松联合胸腺肽α1治疗组(地塞米松0.25 g/kg,胸腺肽5 mg/kg),腹腔注射给药14 d,用卵清蛋白(OVA)致敏建立大鼠哮喘模型,正常组腹腔注射生理盐水做对照,ELISA检测血清及肺泡灌洗液中IL-25和IL-5的水平,Western blot检测肺组织NF-κB的表达情况,以及与IL-25表达水平的相关性分析。结果哮喘组IL-25和IL-5及NF-κB的表达水平均高于其他各组,经地塞米松、胸腺肽α1及二者联合治疗的各组与哮喘组比较均明显下降(P<0.01),血清及肺泡灌洗液IL-25和IL-5的表达水平的相关性分析,结果显示IL-25和IL-5的表达水平成高度正相关(r=0.97,r=0.93,P<0.01),IL-25与NF-κB表达水平成高度正相关(r=0.96,P<0.01),与哮喘组比较胸腺肽α1均能明显降低IL-25、IL-5和NF-κB的表达水平(P<0.01)。结论胸腺肽α1均能通过能够抑制IL-25及IL-5的表达,这种抑制作用可能与NF-κB表达密切相关。
Objective To investigate the effect of thymosin alpha 1 on interleukin-25 expression in asthmatic rats and the mechanism of the regulation of Th2/Th1 balance.Methods Male SPF grade Wistar rats were randomly divided into 5 groups:normal group,asthma model group,dexamethasone treatment group(0.5 mg/kg),thymosin alpha 1 treatment group(5 mg/kg),and dexamethasone combined with thymosin alpha 1 treatment group(0.25 g/kg dexamethasone treatment,5 mg/kg thymosin).Rat asthma model was established by intraperitoneal injection of 14 d,sensitized with ovalbumin(OVA).The normal group was given intraperitoneal injection of saline to compare,and then ELISA was used to detect interleukin-25 and interleukin-5 levels in serum and alveolar lavage fluid(BALF).NF-kappa B expression in lung tissues was detected by Western blotting.The correlation was analyzed between interleukin-25 and interleukin-5 expression.Results Expression levels of interleukin-25,interleukin-5 and the NF-kappa B were higher in asthma group than those in the other groups.Compared with asthmatic group,the dexamethasone group,thymosin alpha 1group and combined groups were significantly decreased(P<0.01).Expression level of IL-25 and IL-5 in serum and alveolar lavage fluid showed a high positive correlation between IL-25 and IL-5 expression levels(r=0.97,r=0.93,P<0.01).IL-25 is highly correlated with NF-κB expression levels(r=0.96,P<0.01).Thymosin alpha1 significantly decreased the expression of IL-25,IL-5 and NF-κB compared with the asthma group.Conclusion Thymosinα1 can inhibit the expression of interleukin-25 and interleukin-5.The mechanism may be closely related to the expression of NF-kappa B.
