摘要
B类基因是调控植物花发育形成的重要因子,分为AP3/DEF和GLO/PI两个亚家族。为了解滇水金凤花发育过程中的分子遗传机制,本研究以滇水金凤花器官为试验材料,通过克隆得到了3个AP3/DEF同源基因,分别命名为IuDEF1、IuDEF2和IuDEF3;对其序列和结构域进行比对分析,发现这3个基因具有高度保守的MADS和K结构域,属于MADS-box家族的TypeⅡ型基因。系统进化分析发现IuDEF1、IuDEF2和IuDEF3可能为旁系同源基因。荧光定量分析表明在早期花器官发育中3个基因的表达量均最高;其空间表达模式显示,在滇水金凤雄蕊中大量表达,萼片和心皮次之,翼瓣中的表达最弱。本研究从分子水平上对滇水金凤花发育有作用的AP3/DEF同源基因进行分析,结果表明AP3/DEF同源基因除了调控萼片和雄蕊的发育外,还可能在心皮发育过程中起着重要作用。
The class B genes play an important role in regulating plant flower development and are divided into AP3/DEF and GLO/PI subfamilies. To understand the molecular genetic mechanism on the flower development of Impatiens uliginosa. In this study, the whole flowers of I. ulignosa were used as experimental materials, three AP3/DEF homologous genes were obtained by cloning, named IuDEF1, IuDEF2 and IuDEF3. Based on analyzing their sequences and domains, 3 genes had the MADS and K typical domains and belong to the Type Ⅱ gene of the MADS-box family. The results of their phylogenetic tree showed that they might belong to paralogous genes. On the basis of analysis of real-time PCR, the expression levels of all three genes were highest in early flower organ development among all tested flower development stages. The results of spatial expression patters showed that a large amount of their expression in the stamens of Impatiens uliginosa was investigated, followed by in the lower sepals and ovary, and their expressions in the lateral petal were the weakest. The above-mentioned results indicated that the AP3/DEF gene not only regulated the development of sepals and stamens, but also played a key role in ovary development.
