摘要
目的:考查盐胁迫对黄蜀葵生长生理及总黄酮含量的影响。方法:采用自吸水耐盐鉴定法,人为设置不同的基质含盐量,以清水为对照,比较6个浓度梯度盐胁迫处理下,黄蜀葵生长生理指标和总黄酮含量的变化规律。利用HPLC法检测不同浓度盐胁迫后黄蜀葵花冠中总黄酮的含量。结果:黄蜀葵为轻度耐盐植物,1~3 g/L盐胁迫能够促进黄蜀葵植株的生长,大于3 g/L后随着盐胁迫浓度增加,其生长受到抑制,在小于9 g/L的盐胁迫下,能完成生活史。采取优化后的超声波提取植物总黄酮,最优超声波提取条件:提取剂为80%的乙醇,提取温度为50℃,提取时间为60 min,料液比为1∶25;盐胁迫浓度为2 g/L时,黄蜀葵花冠中总黄酮含量最高,达5.018%。结论:优化形成了一套高效提取和检测黄蜀葵总黄酮含量的方法,明确了盐胁迫对黄蜀葵生长和总黄酮含量的影响,为黄蜀葵在盐碱区的合理开发利用提供技术支撑和理论基础。
Objective:To investigate the effects of salt stress on growth physiology and total flavonoids content of Abelmoschus manihot.Methods:The self-water absorption and salt tolerance identification method was adopted,the salt content of different substrates was artificially set.The changes of growth physiological indexes and total flavonoids content of Abelmoschus manihot were studied under six concentrations of salt stress treatments with water as contrast.The content of total flavonoids in the Abelmoschus manihot corolla under different salt stress was detected by HPLC method.Results:Abelmoschus manihot was a mildly salt-tolerant plants,and the salt stress of 1~3 g/L could promote the growth of Abelmoschus manihot plants,after more than 3 g/L,the growth was inhibited with the increase of salt stress concentration,but under the salt stress of less than 9 g/L,the whole growth period could be completed.The optimized ultrasonic extraction of total flavonoids from plants was carried out,the optimal ultrasonic extraction condition was as follows:the extraction agent was 80%ethanol,extraction temperature was 50℃,extraction time was 60 min,ratio of material to liquid was 1∶25;When the salt stress concentration was 2 g/L,the total flavonoid content in corolla of Abelmoschus manihot was the highest,reaching 5.018%.Conclusion:A set of methods for efficient extracting and determining the growth and total flavonoids content in the Abelmoschus manihot is optimized and established,the effect of salt stress on the growth and total flavonoids content of Abelmoschus manihot is determined.It provides technical support and theoretical basis for rational development and utilization of Abelmoschus manihot in saline area.
