摘要
试验旨在探究绵羊CARTPT基因所编码蛋白的结构与功能,利用生物信息学软件研究其理化性质、亲疏水性、信号肽、跨膜区域、蛋白质二级结构与三级结构、亚细胞定位和蛋白结构域、糖基化和磷酸化位点,并构建不同物种的CARTPT基因的系统发育树。结果表明:绵羊CARTPT基因CDS区全序列长为351 bp,编码116个氨基酸,编码蛋白的相对分子质量12790.18 Da,理论等电点8.22,脂肪系数114.48,总平均亲水性-0.006,属于分泌蛋白,没有跨膜区域结构;二级结构中主要以α-螺旋与无规则卷曲为主;亚细胞定位分析预测该蛋白最可能在细胞外基质发挥其生物学作用,有1个跨膜结构域;该蛋白质有5个丝氨酸(Ser)、1个苏氨酸(Thr)、2个酪氨酸(Tyr)磷酸化位点;系统进化树发现绵羊与山羊亲缘关系最近。
To explore the sheep CARTPT gene encoding protein structure and function,the test utilized bioinformatics software,researched its physical and chemical properties,personal relationship,signal peptide,transmembrane water area,protein secondary structure and tertiary structure,subcellular localization and protein structure domain,glycosylation and phosphorylation sites,and built the phylogenetic tree CARTPT genes from different species.The results showed that the complete sequence length of the CDS region of sheep CARTPT gene was 351 bp,encoding 116 amino acids,the relative molecular weight of the coding protein was 12790.18 Da,the theoretical isoelectric point was 8.22,the fat coefficient was 114.48,and the total average hydrophilicity was-0.006,which belonged to the secreted protein without the transmembrane regional structure.In the secondary structure,the majority of the structure wasα-helix and irregular curl.Subcellular localization analysis predicted that the protein was most likely to play its biological role in the extracellular matrix and had a transmembrane domain.The protein had five serine(Ser),one threonine(Thr)and two tyrosine(Tyr)phosphorylation sites.The phylogenetic tree found that sheep are the closest relatives of goats.
