摘要
为探明几种不同因素对大孔树脂纯化狼爪瓦松愈伤组织多糖的影响,该试验对大孔吸附树脂种类、进样浓度、进料体积、进料流速和洗脱剂浓度、洗脱剂体积及洗脱流速进行筛选。结果表明:在D101、AB-8、S-8和HP-20等4种大孔树脂中,用AB-8树脂的多糖吸附率和解析率明显好于其它树脂种类;当进样浓度为1.5 mg/mL,进料体积为2 BV,进料流速为1 BV/h时,多糖泄露最少,说明此条件下,树脂可以吸附较多的多糖;在洗脱剂浓度为65%,洗脱剂体积为2.5 BV,洗脱流速为2 BV/h时,最利于多糖的解析。
In order to investigate the effects of several different factors on the purification of polysaccharides from the macroporous resin of Orostachyscartilaginea A.Ber callus,this study investigated the types of macroporous adsorption resin,injection concentration,feed volume,feed flow rate and eluent concentration,washing Filter volume and flow rate.The results showed that,among the four resins of D101,AB-8,S-8,and HP-20,the polysaccharide adsorption rate and resolution rate of AB-8 resin were significantly better than other resin types.When the injection concentration was 1.5 mg/mL,the feed volume was 2 BV,and the feed flow rate was 1 BV/h,the polysaccharide leakage was minimal,indicating that under this condition,the resin could adsorb more polysaccharides.When the concentration of the eluent was 65%,the volume of the eluent was 2.5 BV,and the elution flow rate was 2 BV/h,it was most conducive to the analysis of polysaccharides.
