摘要
为明确宁波口岸从进境日本茶梅Camellia sasanqua种苗花朵上截获的1株疑似山茶叶杯菌Ciborinia camelliae菌株CM-2的分类地位,通过形态学特征观察,ITS、28S、β-微管蛋白基因(β-tubulin)序列比对分析以及致病性测定进行种类鉴定。结果表明:该菌株可在马铃薯葡萄糖琼脂、燕麦琼脂和茶梅花水琼脂培养基平板上生长,但在麦芽汁培养基平板上生长十分缓慢;经培养可产生菌核和小型分生孢子。菌核圆形至椭圆形、浅盘状,大小为(1.86~14.50)mm×(1.37~10.83)mm,成熟时黑色;小型分生孢子球状,直径3.1~4.2μm,易脱落,有时呈短链状。该菌株与山茶叶杯菌菌株EFA-2、EFA-4序列相似度较高,构建的系统发育树中处于同一个分支。该菌株可侵染茶梅,引起山茶花腐症状并产生菌核。结合上述结果,将该菌株鉴定为山茶叶杯菌,这是我国首次截获山茶叶杯菌这一进境检疫性真菌。
The morphological observation,sequence analysis and pathogenicity test were carried out to identify the fungal strain CM-2 isolated from the Camellia sasanqua imported from Japan.The isolate grew well on three kinds of media,including potato dextrose agar(PDA),oatmeal agar(OA)and camellia water agar(CWA),but not well on malt extract agar(MEA).The morphological results showed that the isolate usually produced sparse aerial mycelia and spermatia.The microconidia were globose,3.1-4.2 μm in diameter,usually separating readily but sometimes adhering in short chains.Sclerotia were typically circular to elliptical outline,(1.86-14.5)×(1.37-10.83)mm,at maturity black throughout.Its ITS,28 S and β-tubulin sequences were most similar to those of Ciborinia camelliae(strain EFA-2,EFA-4)deposited in GenBank,phylogenetically conspecific to C.camelliae.The pathogenicity test showed that the strain CM-2 was able to infect Ca.sasanqua by forming sclerotia and cause the symptom of flower blight.The strain was identified as C.camelliae.This is the first report of interception of C.camelliae in China.
