摘要
二孢拟奥德蘑(Hymenopellis raphanipes)是一种新型食用菌,目前对其研究较少。为进一步研究二孢拟奥德蘑,通过在前期单因素试验筛选二孢拟奥德蘑母种培养基中的碳源、氮源、氨基酸、维生素、无机盐等结果的基础上,选取淀粉、麦芽浸膏、酵母膏、VB1、天冬氨酸、脯氨酸、硫酸铵、碳酸钙清液、磷酸二氢钾、硫酸镁等10个因子,采用U12(1210)均匀设计表设计试验方案,优化二孢拟奥德蘑母种培养基,以获得二孢拟奥德蘑最佳母种培养基配方。结果表明,二孢拟奥德蘑菌丝生长最适培养基为每升培养基中含淀粉27.50 g、酵母膏5.50 g、硫酸铵1.10 g、天冬氨酸0.02 g,脯氨酸220.00μg、VB11.44μg,可溶性碳酸钙清液11.00 g、磷酸二氢钾10.97 g,该最佳配方有利于其菌丝生长,优于PDA培养基。
Hymenopellis raphanipes is a new type of edible fungi,but there were few researches on it at present.In order to further study H.raphanipes,starch,malt extract,yeast extract,VB1,aspartic acid,proline,ammonium sulfate,calcium carbonate clear liquid,potassium dihydrogen phosphate and magnesium sulfate were selected as ten test factors based on the results of screening the carbon source,nitrogen source,amino acid,vitamin,inorganic salt and other ingredient results in the stock medium of H.raphanipes by single factor test in the early stage.Each factor was divided into 12 levels.U12(1210)uniform design table was used to arrange the experiment scheme and to optimize the stock medium of H.raphanipes.The results showed that the optimum stock medium of H.raphanipes was starch 27.50 g,yeast extract 5.50 g,ammonium sulfate 1.10 g,aspartic acid 0.02 g,proline 220.00μg,VB11.44μg,soluble calcium carbonate clear liquid 11.00 g and potassium dihydrogen phosphate 10.97 g.The mycelia of H.raphanipes grew faster in the optimum stock medium than in PDA medium,and the mycelia was more dense and white.
