摘要
目的:运用网络药理学探讨牛膝治疗骨关节炎有效成分的作用靶点及通路。方法:通过TCMSP数据库挖掘牛膝的有效成分并预测其作用靶点,利用GeneCards、OMIM等数据库筛选骨关节炎的靶标,运用Cytoscape软件构建药物-活性成分-靶点的关系网络,通过STRING数据库构建靶点蛋白-蛋白相互作用网络,并对关键靶点的生物功能注释及通路进行分析。结果:预测得到20个牛膝治疗骨关节炎的有效成分和47个有效靶点,并推测其作用机制可能与细胞凋亡信号通路、多种物种-细胞凋亡信号通路、肿瘤坏死因子信号通路、白细胞介素-17信号通路等有关,且前列腺素G/H合成酶1、雄激素受体、雌激素受体1等靶点基因可能起关键性作用。结论:牛膝有效成分可能通过调控炎症因子及细胞凋亡等药理作用发挥治疗骨关节炎的作用。
Objective:To explore the target and pathway of active ingredients from Niuxi(Radix Achyranthis Bidentatae)for osteoarthritis by network pharmacology.Methods:TCMSP database was used to mine the effective components of Niuxi and predict their target.Databases such as Genecards and OMIM were used to screen the targets of osteoarthritis.The relationship network of drug,active components and target was constructed by using software Cytoscape.The interaction network of target protein was constructed by STRING database.The biological function annotation and pathway of key targets were analyzed.Results:Twenty active components and 47 effective targets of Niuxi for osteoarthritis were predicted,and the mechanism of action might be related to apoptosis signaling pathway,multi species apoptosis signaling pathway,tumor necrosis factor signaling pathway,interleukin-17 signaling pathway,and prostaglandin G/H synthetase 1,androgen receptor,estrogen receptor 1and other target genes may play a key role.Conclusion:The effective components of Niuxi may play a role in the treatment of osteoarthritis by regulating inflammatory factors and apoptosis.
