朝仓花椒转录组密码子偏好性分析 被引量:12

Preference Analysis of Transcriptome Codon in Zanthoxylum piperitum DC.var.inerme Makino

摘要 生物体内密码子的使用偏好性对外源基因在宿主内表达存在较大的影响。为研究朝仓花椒密码子偏好性,本研究利用CodonW软件对花椒7 014个编码氨基酸基因序列进行密码子偏好性分析,计算了编码氨基酸的密码子使用频率,结果显示花椒偏好使用以碱基A/U结尾的密码子,共有23个,如AGA、CCU、CCA等。少量使用以碱基G/C结尾的密码子,共有8个,如GAG、UUG、CUC等。与毛果杨、拟南芥、大肠杆菌、酵母、克莱门柚进行密码子偏好性比较,发现花椒与其他物种偏好性差异较大的密码子分别有21、20、26、22和27种,其中与拟南芥差异最小,与大肠杆菌、克莱门柚差异最大。该研究对花椒基因的异源表达及蛋白功能验证具有重要的指导意义。 The use of codons in organisms has a greater impact on the expression of foreign genes in the host. In order to study the coke preference of Zanthoxylum piperitum DC. var. inerme Makino, in this study, CodonW software was used to analyze the codon bias of 7 014 encoded amino acid gene sequences of Zanthoxylum. The frequency of codon usage of the encoded amino acids was calculated. The results showed that Zanthoxylum prefers to use codons ending in base A/U, a total of 23 such as AGA, CCU, CCA, etc. A small number of codons ending in the base G/C are used in a small amount, such as GAG, UUG, CUC, and the like. Compared with Populus trichocarpa, Arabidopsis thaliana, Escherichia coli, Saccharomyces cerevisiae and Citrus clementina, the codon preference was compared. It was found that there were 21, 20, 26, 22 and 27 codons with different preferences for Zanthoxylum and other species. Among them, the difference from Arabidopsis is the smallest, and the difference is the largest with Escherichia coli and Citrus clementina. This study has important guiding significance for the heterologous expression of the Zanthoxylum gene and the verification of protein function.
