蜈蚣败毒饮对银屑病小鼠皮损中Th17细胞主导通路相关因子基因表达影响的实验研究 被引量:9

Experimental Study on the Effect of Wugong Baidu Decoction on the Related Genes Expression of Th17 Cell Dominant Pathway in the Lesions of Psoriasis Mice

摘要 目的观察中药蜈蚣败毒饮对银屑病转基因小鼠模型皮损中Th17细胞主导通路IL-23-STAT3/RORγt-IL-17相关基因表达的影响。方法将银屑病小鼠随机分为空白对照组、甲氨蝶呤对照组、蜈蚣败毒饮低、中、高剂量组,分别进行药液配置、小鼠灌胃、皮损取材,通过皮损组织中RNA提取、引物合成、逆转录及PCR定量分析等对小鼠皮损中IL-17A、IL-23、STAT3、RORγt基因的表达进行检测与分析。结果蜈蚣败毒饮高剂量组和甲氨蝶呤组能够显著降低银屑病小鼠模型皮损中IL-17A、IL-23、STAT3、RORγt的表达(P<0.01),蜈蚣败毒饮中剂量组能够降低皮损中IL-23、STAT3、RORγt的表达(P<0.05),且蜈蚣败毒饮的干预作用随浓度呈现出一定的量效依赖关系。结论中药蜈蚣败毒饮可以通过干预小鼠皮损中Th17细胞主导通路的上游刺激因子IL-23、转录因子STAT3和RORγt、下游分泌因子IL-17A相关基因的表达,从而发挥治疗银屑病的作用。 Objective To observe the effect of traditional Chinese medicine named Wugong Baidu Decoction on the related genes expression of IL-23-STAT3/RORγt-IL-17 that the Th17 cell dominate pathway in the lesions of psoriasis transgenic mice.Methods Psoriasis mice were randomly divided into blank control group, methotrexate control group, the low, medium and high dose groups of Wugong Baidu Decoction, pharmaceutical liquid configuration, gastric perfusion and skin lesions sampling were carried out respectively, detecting and analyzing the genes expression of IL-17 A, IL-23, STAT3 and RORγt in mice skin lesions through methods of RNA extraction, primer synthesis, reverse transcription and quantitative analysis by PCR in skin lesions.Results The expressions of IL-17 A, IL-23, STAT3 and RORγt in the lesions of psoriatic mice model are significantly decreased in the high dose group of Wugong Baidu Decoction and methotrexate group(P<0.01), and the expressions of IL-23, STAT3 and RORγt in skin lesions are decreased in the medium dose group of Wugong Baidu Decoction(P<0.05), and it shows a certain dose-effect dependence relationship of the intervention effect of Wugong Baidu Decoction with the concentration. Conclusion Traditional Chinese medicine Wugong Baidu Decoction can play a role in the treatment of psoriasis by interfering with the expression of related genes of upstream stimulator IL-23, transcription factors STAT3 and RORγt, downstream secretion factor IL-17 A of Th17 cell dominant pathway.
