补肾促排卵汤对多囊卵巢综合征模型大鼠卵巢MMP-2、α-SMA的影响 被引量:7

Effect of Bushen Cupailuan Decoction(补肾促排卵汤)on MMP-2 and a-SMA in the Ovaries of Rats with Polycystic Ovary Syndrome

摘要 目的:探讨补肾促排卵汤对硫酸脱氢表雄酮(dehydroepiandrosterone,DHEA)诱导的多囊卵巢综合征(polycystic ovary syndrome,PCOS)模型大鼠的疗效及对大鼠卵巢MMP-2、α-SMA表达的影响。方法:将50只SD雌性大鼠随机分为空白组、模型组和补肾促排卵汤低、中、高剂量组,每组10只。空白组每日颈部皮下注射0.2 mL大豆油,其余各组每日颈部皮下注射DHEA 60 mg/kg+0.2 mL大豆油,持续注射35 d。造模第21天开始于每日上午制作大鼠阴道分泌物涂片,通过显微镜下观察大鼠阴道分泌物中细胞形态从而判断大鼠的动情周期,大鼠失去规律的动情周期即为造模成功。造模第36天开始连续每天灌胃给药,持续4周。检测阴道分泌物的细胞形态;检测大鼠卵巢中卵泡发育的情况;检测大鼠血清中性激素T、E2、LH、FSH含量的变化;RT-qPCR法检测大鼠卵巢中MMP-2 mRNA、α-SMA mRNA的表达变化;Western blotting检测大鼠卵巢中MMP-2、α-SMA蛋白的表达变化。结果:与空白组比较,模型组大鼠丧失规律的动情周期,大鼠卵巢中卵泡呈囊状扩张,血清T、LH水平明显升高(P<0.01),同时MMP-2 mRNA及蛋白表达减少(P<0.01),α-SMA mRNA及蛋白表达增多(P<0.01)。与模型组比较,低剂量补肾促排卵汤可改善PCOS模型大鼠动情周期,降低血清LH水平(P<0.05),改善卵泡发育,减少囊状扩张卵泡的形成,上调卵巢MMP-2 mRNA及蛋白的表达(P<0.05);中、高剂量补肾促排卵汤可明显改善PCOS模型大鼠动情周期,降低血清T、LH水平(P<0.05或P<0.01),改善卵泡发育,减少囊状扩张卵泡的形成,上调卵巢MMP-2mRNA及蛋白的表达(P<0.01),下调α-SMA mRNA及蛋白的表达(P<0.05或P<0.01)。结论:补肾促排卵汤能有效治疗DHEA诱导的PCOS模型大鼠,与调控大鼠卵巢中MMP-2、α-SMA的分子表达相关。 Objective:To investigate the therapeutic effect of Bushen Cupailuan Decoction on polycystic ovary syndrome(PCOS)model rats induced by dehydroepiandrosterone sulfate(DHEA)and the expression of MMP-2 and a-SMA in ovaries of rats.Methods:A total of 50 SD female rats were randomly divided into blank group,model group and Bushen Cupailuan Decoction low,medium and high-dose groups,with 10 rats in each group.The blank group was injected with 0.2 mL of soybean oil subcutaneously in the neck every day,and the other groups were injected with DHEA 60 mg/kg+0.2 mL of soybean oil subcutaneously in the neck every day for 35 days.On the 21 stday of modeling,a smear of rat vaginal secretions was made every morning,and the estrus cycle of rats was judged by observing the cell morphology of rat vaginal secretions under a microscope.Rats losing their regular estrus cycle was established successfully.On the 36 thday of modeling,the drug was given by gavage every day for 4 weeks.The cell morphology of vaginal secretions,follicular development in ovaries and the changes of serum sex hormones T,E2,LH and FSH were detected.RT-qPCR was used to detect the expression of MMP-2 andα-SMA mRNA in the ovaries of rats.The expression of MMP-2 andα-SMA in the ovaries of rats was detected by Western blotting.Results:Compared with the blank group,the rats in the model group lost the regular estrous cycle,the follicles in the ovaries showed cystic expansion,the levels of serum T and LH were significantly increased(P<0.01),the expression of MMP-2 mRNA and protein was decreased(P<0.01),and the expression ofα-SMA mRNA and protein was increased(P<0.01).Compared with the model group,low dose of Bushen Cupailuan Decoction could improve the estrous cycle,decrease the serum LH level(P<0.05),improve the follicular development,reduce the formation of cystic expanded follicle,and up regulate the expression of MMP-2 mRNA and protein in the ovary(P<0.05).Medium and high-dose of Bushen Cupailuan Decoction could significantly improve the estrous cycle of PCOS model rats,reduce serum T and LH levels(P<0.05 or P<0.01),improve follicular development,reduce the formation of cystic expanded follicles,up-regulate the expression of MMP-2 mRNA and protein(P<0.01),and down-regulate the expression ofα-SMA mRNA and protein(P<0.05 or P<0.01).Conclusion:Bushen Cupailuan Decoction can effectively treat PCOS model rats induced by DHEA,which is related to the regulation of the molecular expression of MMP-2 andα-SMA in the ovaries of rats.
